低氧诱导因子-1α在糖尿病肾病中的表达及其意义 被引量:2

Expression and Significance of HIF-1α in Kidney Tissue of Rats with Diabetic Nephropathy

摘要 目的探讨低氧诱导因子-1α(HIF-1α)在实验性糖尿病肾病中的表达。方法将30只健康Wister大鼠适应性喂养1周后随机分为模型组和对照组各15只,其中模型组于左下腹腔注射链脲佐菌素制备糖尿病模型,对照组腹腔注射等量的柠檬酸缓冲液。两组均自心脏取血,采用全自动生化分析仪测定血清肌酐(Scr)、尿素氮(BUN),并计算内生肌酐清除率(Ccr);采用免疫组化及westerblot分析法测定肾脏组织中HIF-1α表达。结果模型组血清BUN显著高于对照组,Ccr显著低于对照组,HIF-1α表达显著高于对照组。结论 HIF-1α在糖尿病肾病发生、发展中起重要作用,此为糖尿病肾病防治及预后判定提供了依据。 Objective To investigate the expression and significance of hypoxia inducible factor 1 alpha(HIF-1 alpha) in experimental diabetic nephropathy.Methods Diabetes mellitus was induced in male Wister rats by an intraperitoneal injection of streptozotocin(60mg/kg).The serum biochemical parameters in rats were measured by automatic chemistry analyzer.The expression of HIF-1α protein was examined by Western blot and immunohistochemistry.Results In the model group,the serum BUN was significantly higher than that of control group,creatinine clearance(Ccr) was significantly lower than that of control group.The expression of HIF-1α protein was significantly higher than control group.Conclusion The expression of HIF-1α protein is upregulated in diabetic nephropathy.HIF-1α protein is a promising new therapeutic target for treating diabetic nephropathy.
