口腔上皮细胞感染白假丝酵母菌后HBD-2蛋白的表达 被引量:1

摘要 目的检测人β防御素2(Human beta defensin-2,HBD-2)在口腔上皮细胞中的表达以及白假丝酵母菌对其表达的影响。方法分别以孢子相、菌丝相以及灭活的白假丝酵母菌与KB细胞共同培养6、12、24h后,采用ELISA技术检测各组KB细胞培养液中HBD-2蛋白的表达情况,与对照组进行比较。结果 KB细胞有HBD-2蛋白的基础表达,感染6h后,各实验组与对照组之间表达差异无统计学意义(P>0.05);感染12h后,孢子组(93.974±6.426)、菌丝组(72.287±2.839)显著高于对照组(37.380±5.663),P<0.01;感染24h后,各实验组表达均高于对照组(P<0.05),但和12h时的表达相比,孢子组(80.851±7.860)、菌丝组(63.932±8.070)出现了下降趋势(P<0.05)。结论 HBD-2在口腔上皮细胞中有基础表达,白假丝酵母菌能诱导其表达上调;白假丝酵母菌对HBD-2的诱导表达只发生在感染的特定时期内,当菌丝进一步侵袭后,HBD-2的表达会逐渐减弱。 Objective To determine the expression of Human beta Defense-2(HBD-2) in oral epithelial cells and to analyze the influence of C.albicans infection on its expression.Methods KB cells was infected with blastospores phase,hyphae phase,and inactivated C.albicans for 6,12,24 hours.Expression of HBD-2 in each group was determined by enzyme-linked immunosorbent assay(ELISA).Results HBD-2 was basally expressed in KB cells.After 6 hours of infection,the expression showed no significant difference(P〉0.05) between experiment groups and control group;After 12 hours of infection,the expression showed significant upregulation in the blastospores group(93.974±6.426)and hyphae group(72.287±2.839) compared to the controls(37.380±5.663),P〈0.01;After 24 hours of infection,the expression of HBD-2 in experiment group was upregulation compared to the controls(P〈0.05),but it was found decrease in the blastospores group(80.851±7.860)and hypha group(63.932±8.070),P〈0.05.Conclusion HBD-2 was basally expressed in oral epithelial cells.C.albicans can upregulate the expression levels;The expression of HBD-2 was induced after a certain period of infection with C.albicans,but the expression regressed as hyphal invasion progressed.
