应用Fluoro-Jade C评价小鼠匹罗卡品模型中岛皮质神经变性

摘要 目的应用Fluoro-Jade C(FJC)染色方法在小鼠匹罗卡品癫痫模型中探测岛皮质结构中神经元的变性情况,以了解岛皮质结构在慢性颞叶癫痫发生中的病理变化和癫痫反复发作的神经基础。方法雄性昆明小鼠6只随机分为对照组3只和匹罗卡品处理组3只。对照组给予阿托品、生理盐水腹腔注射;匹罗卡品处理组给予阿托品、匹罗卡品腹腔注射。在癫痫持续状态后3 d通过经心灌注固定处死匹罗卡品处理组小鼠。对照组小鼠处死时间及方法同匹罗卡品处理组。在岛皮质水平切制冠状切片,行FJC染色,在荧光显微镜下观察FJC阳性细胞的形态和在岛皮质中的分布情况。结果匹罗卡品处理组FJC染色的脑切片上可清楚看到岛皮质出现亮黄绿色荧光的FJC阳性细胞,呈神经元形态,胞体和突起均清晰显示。而对照组未见此显示。结论 FJC染色技术在小鼠匹罗卡品癫痫模型中显示岛皮质发生了神经元变性,有利于更好地理解颞叶癫痫中所发生的病理变化和癫痫反复发作的发生机制。 Objective To detect degenerative neurons by Fluoro-Jade C(FJC) that may occur in the insular cortex of pilocarpine-treated mice and to understand the neural basis of insular cortex in long-term brain pathological changes and recurrent seizure mechanism of chronic epilepsy.Methods Six Kunming male adult mice were divided into pilocarpine-treated group(n=3) and control group(n=3).The mice in control group were injected intraperitoneal with atropine and saline.The pilocarpine-treated mice were injected intraperitoneal with atropine and pilocarpine.Pilocarpine-treated mice were sacrificed at 3 days after status epilepticus.Control mice were also sacrificed at 3 days.All the pilocarpine-treated and control mice were sacrificed by perfusion fixation.Brain coronal sections in the insular cortex levels were subject to FJC staining.Finally,FJC-stained brain sections were examined under an epifluorescence microscope to identify the shape and overall distribution of the FJC positive cells in the insular cortex.Results In the brain sections of the pilocarpine-treated group,the FJC-positive staining cells showed a bright green color and with the form of neuron,the soma and apophysis were clearly display.They were not detected in control group.Conclusion This study successfully applied FJC staining to demonstrate neurodegeneration in the insular cortex of pilocarpine-treated mice,which will benefit further understanding the mechanism of brain pathological changes and recurrent seizure in temporal lobe epilepsy.
