贝加灵喷雾剂对大鼠局灶性脑缺血损伤的保护作用 被引量:2

Protective Effect of Baicalin Spray on Focal Cerebral Ischemia in Rats

摘要 目的:研究贝加灵喷雾剂对大鼠局灶性脑缺血损伤的保护作用,并初步探讨其作用机制。方法:SD大鼠随机分为假手术组、模型组、贝加灵高、中、低剂量组、尼莫地平组、空白乳剂组。贝加灵高、中、低剂量组(2.66,1.33,0.66 mg.kg-1)鼻腔给药,尼莫地平组(0.4 mg.kg-1)尾静脉注射给药,5 d后,采用血管内栓线阻断法建立大鼠局灶性脑缺血模型,继续给药2 d,观察大鼠神经功能缺损情况,测定各组大鼠脑组织含水量,利用紫外分光光度法测定大鼠局灶性缺血脑损伤血清中超氧歧化酶(SOD)活力、脑组织中Ca2+-ATP酶含量。结果:与模型组比较,贝加灵高、中、低剂量组神经功能评分(1.06±0.80),(1.00±0.84),(0.89±0.94)分,极显著性降低(P<0.01);高、中剂量组脑组织含水量(79.33±1.46)%,(79.34±1.01)%,极显著性降低(P<0.01),低剂量组含水量(79.72±1.87)%显著性降低(P<0.05);血清中SOD活力(177.622±5.224),(160.350±41.139),(167.279±6.450)U.mL-1,脑组织Ca2+-ATP酶含量(3.444±3.730),(3.192±1.298),(2.866±0.156)U.mg-1,均显著性降低(P<0.05)。结论:贝加灵喷雾剂对大鼠局灶性脑缺血损伤有一定修复保护作用,可能通过抗自由基和改善能量代谢发挥这一作用。 Objective: To study the protective effect of baicalin spray on focal cerebral ischemia in rats,and to explore its possible therapeutic mechaniam.Method: SD rats were randomly divided into seven groups: sham operation group,model group,baicalin's spray groups with high,middle and low dose,nimodipine 0.4 mg·kg-1 group and blank emulsion group.Baicalin's spray groups were given baicalin(2.66,1.33,0.66 mg·kg-1,respectively)by nasal administration,and nimodipine group was injected.After 5 days,focal cerebral ischemia model was established by middle cerebral artery occlusion(MCAO).After 2 days of continued administration,the extent of neurological deficits was evaluated by Longa method and the content of brain water was assayed by weighing method,the level of SOD in serum and Ca2+-ATP in brain tissues were determined by the Ultravident Spectrophoto-metric.Result: Compared with the model group,neurological deficits scores of baicalin's spray groups with high,middle,low dose group(1.06±0.80),(1.00±0.84),(0.89±0.94),was significantly decreased(P0.01);the content of brain water(79.33±1.46)%,(79.34±1.01)% was significantly decreased(P0.01),the low dose groups was also decreased(P0.05).The content of SOD in serum(177.622±5.224) U·mL-1),(160.350±41.139) U·mL-1,(167.279±6.450) U·mL-1,and the content of Ca2+-ATP in brain tissues(3.444±3.730),(3.192±1.298),(2.866±0.156) U·mg-1 were highly increased(P0.05).Conclusion: Baicalin spray has protective effect on focal cerebral ischemia injury;its machanism may be related to the anti-free radical and improvement of energy metabolism.
