摘要
目的用超高效液相色谱法测定波棱瓜子中Herpetrione的含量。方法采用Waters ACQUITY UPLC BEHC18色谱柱(2.1 mm×50 mm,1.7μm),流动相为乙腈-0.1%磷酸溶液(21∶79),流速为0.5 ml/min,柱温:30℃,检测波长:240 nm。结果 Herpetrione在0.0095～0.1520μg范围内线性关系良好,其回归方程为:Y=2.68×106X-278.98,r=0.9999;平均回收率为99.57%,RSD为1.15%。结论该方法操作简单,准确性和重复性好,建立了波棱瓜子的一种快速准确的含量检测方法。
Objective To establish a method for determining the content of herpetrione in Herpetospermum caudigerum by ultra high performance liquid chromatography. Methods Waters ACQUITY uPLc BEH C18 chromatographic column （2.1 mm× 50 mm, 1.7 μm） was used. The acetonitrile-0.1% phosphonic acid solution （21：79） was used as the mobile phase. The flow rate, column temperature and detection wavelength were set at 0.5 ml/min, 30℃ and 240 nm respectively for determination. Results In the range of 0. 0095-0. 1520 p,g, herpetrione showed good linear relationship and its regression equation was Y = 2.68 ×106X - 278.98, r = 0. 9999. The average recovery of this determination was 99.57% and its RSD was 1.15%. Conclusion This method is user-friendly, and precise and reproducibly. It is suitable for quality control of Herpetospermum caudigerum.
