摘要
目的建立测定大鼠血清中柔红霉素血药浓度的高效液相色谱法。方法色谱柱为Phenomenex C18柱(250 mm×4.6 mm,5μm),流动相为甲醇-0.01 mol/L磷酸二氢铵溶液(pH=4.65)-冰醋酸(30∶20∶0.1),流速1.0 mL/min,检测波长233 nm,柱温35℃,内标为盐酸多柔比星,进样量20μL。结果盐酸柔红霉素质量浓度在0.147 8~14.77 8 mg/L(r=0.998 4)范围内与待测物和内标峰高比值线性关系良好,低、中、高3种质量浓度(0.738 9,3.694 5,14.778 0 mg/L)的平均相对回收率分别为102.5%,98.7%,99.3%,日内、日间RSD均小于10%。结论该方法样品处理简便,可用于血清中盐酸柔红霉素的血药浓度测定和药代动力学研究。
Objective To establish the determination method of serum concentration of daunorubicin hydrochloride by HPLC.Methods The samples were separated on the Phenomenex C18 column(250 mm×4.6 mm,5 μm) with the mobile phase consisted of methanol-0.01 mol/L ammonium dihydrogen phosphate solution(pH=4.65)-glacial acetic acid(30 ∶20 ∶0.1) at the flow rate of 1.0 mL/min.The UV detection wavelength was 233 nm.The column temperature was 35℃.The internal standard was doxorubicin hydrochloride.The injection volume was 20 μL.Results The linear range for daunorubicin hydrochloride was 0.147 8-14.77 8 mg/L(r=0.998 4).The mean relative recovery rates of daunorubicin hydrochloride at low,middle and high concentrations(0.738 9,3.694 5,14.778 0 mg/L)were 102.5%,98.7%and 99.3%,respectively.Both intra-day and inter-day RSD were below 10%.Conclusion This method is simple and suitable for blood concentration monitoring and pharmacokinetic study of daunorubicin hydrochloride.
