摘要
目的:研究白芷醇提物延缓皮肤衰老与抗氧化作用的相关性,探讨其延缓皮肤衰老可能的机制。方法:分别以1,1-二苯基-2-三硝基苯肼自由基(DPPH.)、超氧阴离子自由基(O2-)、羟自由基(OH.)3种自由基体系对白芷醇提物进行体外抗氧化活性测定。采用D-半乳糖颈背部sc复制小鼠亚急性衰老模型,白芷醇提物设高、中、低剂量(6.0、3.0、1.5mg.10g-1)组,连续ig给药42d后,对小鼠皮肤匀浆中3种抗氧化指标和2种皮肤衰老化指标进行测定,并对抗氧化指标与皮肤老化指标进行相关性分析。结果:白芷醇提物体外对3种自由基均有较强的清除作用,且存在一定的量效关系。高、中剂量白芷醇提物均能显著提高衰老模型小鼠皮肤中超氧化物歧化酶(SOD)活性,降低丙二醛(MDA)和脂褐素(LF)含量,并能显著提高皮肤中羟脯氨酸(Hyp)和水分的含量(P<0.01或P<0.05);低剂量白芷醇提物能显著提高SOD活性,降低MDA含量,并显著提高皮肤含水量(P<0.05)。高剂量白芷醇提物对Hyp的影响与对SOD、MDA、LF等抗氧化指标的影响有较密切相关性(r=0.921)。结论:白芷醇提物具有较强的体内、外抗氧化活性,这一活性与其延缓皮肤衰老作用密切相关,且可能是其美白抗衰老作用机制之一。
OBJECTIVE:To investigate the relationship between the anti-aging function and antioxidation ethanol extract from Angelicae Dahuricae Radix.METHODS:Firstly,we study the antioxidant activities of ethanol entract from Angelicoe Dahuricae Radix in vitro based on DPPH·free radical,O2-free radical,OH·free radical,respectively.Secondly,the sub-acute aging model mice were established by subcutaneous injection of D-galactose in the napes and backs.The mice of ethanol extract from Angelicae Dahuricae Radix medication groups were fed ethanol extract from Angelicae Dahuricae Radix with three doses(6.0,3.0,1.5 mg·10 g-1) intragastrically for 42 days.3 antioxidation index and 2 skin aging index in mice skin homogenate were determined,and correlation analysis of them was conducted.RESULTS:Ethanol extract from Angelicae Dahuricae Radix showed significant clearance effects on 3 free radicals in concentration-dependent manner.Also,the high and medium doses of the extract could significantly increase the activity of SOD,reduce the content of MDA and lipofuscin(LF)in the skin of the mice,meanwhile,they could significantly increase the content of hydroxyproline(Hyp)and moisture(P0.01 or P0.05);the low dose could increase the activity of SOD,reduce the content of MDA,and could increase the content of moisture in the mice skin(P0.05).There was a good correlation between the effects of high dose of ethanol extract from Angelicae Dahuricae Radix on Hyp and that of oxidative index such as SOD,MDA,LF(determining coefficient r=0.921).CONCLUSION:Ethanol extract from Angelicae Dahuricae Radix has strong antioxidative effects both in vitro and in vivo.There is a good correlation between its antioxidative function and its anti-aging function to hint that its antioxidative activity could be one of the whitening anti-aging mechanisms.
