乙醇浸提法提取槐角愈伤组织中异黄酮的研究 被引量:5

摘要 以国槐槐角培养出的愈伤组织为试材,以槐角苷为标准品,利用紫外可见光分光光度法研究乙醇浸提法提取国槐槐角愈伤组织中异黄酮的工艺条件。在单因素试验的基础上,设计了L9(34)(4因素3水平)的正交试验。研究结果表明:乙醇浸提法提取槐角异黄酮的最佳工艺条件为:提取温度75℃;提取时间9 h;提取乙醇浓度60%;提取的液料比70∶1,槐角异黄酮的最大提取量为:12.045 mg/g。 In this paper, the contents of total isoflavone in callus of Sophora japonica L. Fruits were determined by violet and visible spectrophotometric methods in terms of sophoricoside equivalent. First of all, the single factor experiments were conducted, then the orthogonal experiment of L9 (34) (four factors three levels) by colorimetric estimation was designed. The results of the study showed that the optimum process of ethanol extraction of isoflavone of Sophora japonica L. Fruits was obtained, when used 75 ℃ extraction temperature,9 h extraction times ,60% ethanol concentration, and extracting liquid and extraction agent ratio was 70: 1. The extraction capacity of isoflavone of Sophora japonica L. was 12. 045 mg/g at most.
