补肾、健脾、活血中药复方对糖皮质激素性骨质疏松症大鼠肾组织Runx2 mRNA及蛋白表达影响的比较研究 被引量:6

摘要 目的:观察糖皮质激素性骨质疏松症(GIOP)大鼠肾组织Runx2 mRNA及蛋白表达,探讨GIOP的发病机制及补肾中药复方的疗效机理,并与健脾、活血中药复方比较。方法:肌注地塞米松复制GIOP大鼠模型,实验设正常组、模型空白组、补肾中药复方组、健脾中药复方组、活血中药复方组、骨疏康颗粒阳性对照组。造模及灌胃给药9周。应用XR-26型双能X线骨密度仪测定股骨骨密度,实时定量RT-PCR及Western Blot检测肾组织Runx2 mRNA及蛋白表达。结果:(1)股骨骨密度:与正常组比较,模型空白组明显降低(P<0.01);与模型空白组比较,补肾中药复方组明显升高(P<0.01),骨疏康颗粒阳性对照组明显升高(P<0.05),健脾中药复方组和活血中药复方组虽有升高趋势,但无统计学意义(P>0.05)。(2)肾组织Runx2 mRNA及蛋白表达:与正常组比较,模型空白组明显升高(P<0.01);与模型空白组比较,补肾中药复方组明显下调(P<0.01),骨疏康颗粒阳性对照组明显下调(P<0.05),健脾中药复方组和活血中药复方组虽有所降低,但无统计学意义(P>0.05)。结论:肾组织Runx2 mRNA及蛋白表达升高可能是GIOP的发病机制之一;补肾中药复方可能通过下调肾组织Runx2mRNA及蛋白表达,有效防治GIOP,其作用优于健脾、活血中药复方。 Objective:To observe mRNA and protein expression of Runx2 in kidney tissues of the rats with glucocorticoid-induced osteoporosis(GIOP),discuss the pathogenesis of GIOP and the effective mechanism of reinforcing the kidney herbs compound and compare the therapeutic effect and adjustable function of it with strengthening the spleen herbs compound and promoting blood circulation herbs compound.Methods: We established the GIOP rat models by intramuscular injection of dexamethasone.We divided all the rats into normal group,blank model group,reinforcing the kidney herbs compound group,strengthening the spleen herbs compound,promoting blood circulation herbs compound group and GUSHUKANG granula positive control group.After 9 weeks of establishing models and administrating by gavage,we detected femur bone mineral density by using the XR-26 dual energy X-ray absorptiometry.We detected mRNA expression of Runx2 in kidney tissues by Real-Time Quantitative RT-PCR and detected their protein expression by Western Blot.Results:(1) femur bone mineral density: Compared with normal group,blank model group decreased significantly(P0.01);Compared with blank model group,reinforcing the kidney herbs compound group increased significantly(P0.01),GUSHUKANG granula positive control group increased significantly(P0.05),strengthening the spleen herbs compound group and promoting blood circulation herbs compound group both tended to increase,but neither of their difference had statistical significance(P0.05).(2) mRNA and protein expression of Runx2 in kidney tissues: Compared with normal group,blank model group increased significantly(P0.01);Compared with blank model group,reinforcing the kidney herbs compound group down-regulated significantly(P0.01),GUSHUKANG granula positive control group down-regulated significantly(P0.05),strengthening the spleen herbs compound group and promoting blood circulation herbs compound group both tended to decrease,but neither of their difference had statistic
