摘要
目的通过研究去势骨质疏松小鼠骨髓间充质干细胞(BMMSC)增殖及分化功能的改变,揭示其在骨质疏松发病中的作用。方法建立去势小鼠骨质疏松模型,检测BMMSC增殖和多向分化能力变化,采用反转录酶-聚合酶链反应(RT-PCR)和小核糖核酸(microRNA)芯片对骨质疏松BMMSC的分化相关基因和microRNA表达谱进行检测。结果在雌激素缺乏所致的骨质疏松中,BMMSC的增殖能力和成骨分化能力出现了一定的下降,而成脂分化能力却明显增强,同时发现一组分化功能相关基因及microRNA表达出现改变。结论 BMMSC的功能缺陷与骨质疏松的病理改变相一致,可能在骨质疏松发生中起到重要作用。而部分功能基因及microRNA的改变可能是导致BMMSC功能异常的分子机制。
Objective To investigate the alteration of proliferation and differentiation function of marrow - derived mesenehymal stem cells and to find out its role in the osteoporosis. Methods The proliferation and differ- entiation function of marrow-derived mesenchymal stem cells from mice model of osteoporosis were evaluated, and the differentiation associated gene and microRNA expression profile were detected by RT-PCR and microRNA microarray chip. Results The ability of proliferation and osteogenic differentiation decreased and the ability of adipogenesis differentiation increased in the marrow-derived mesenchymal stem ceils of mice with osteoporosis. A serials of differ- entiation associated gene and microRNA expression were differentially expressed in the osteoporosis marrow-derived mesenchymal stem cells. Conclusion The dysfunction of marrow-derived mesenchymal stem cells is coincident with the pathological expression of osteoporosis and it may play an important role in the osteoporosis. The alteration of the expression of some functional gene and microRNA may be the molecular mechanism of dysfunction of marrow- derived mesenchymal stem cells.
