摘要
[目的]探索提高花粉管通道法对大白菜转化率的途径。[方法]以大白菜品种03号和玉青的花粉为材料,对其萌发的缓冲液和超声波处理的各参数进行筛选。[结果]大白菜花粉萌发的最佳缓冲液为蔗糖200 g/L+硼酸100 mg/L+硝酸钙200 mg/L,较理想的超声波处理参数为功率150 W、工作时间5s、间隔时间5s、处理次数8s;经浓度为200 mg/L的卡那霉素筛选,获得3株T1代单株。[结论]为进一步进行大白菜的基因转化奠定了基础。
[Objective] The Research aimed to explore improving the percent conversion of Chinese cabbage through pollen tube pathway.[Method] The Chinese cabbage variety: Dabaicai No.03 and Yuqing being taken as experimental material,the parameters of the germination pollen treated by buffer and ultrasonic was analyzed.[Results] The best buffer for cabbage pollen germination was the sucrose of 200 g/L + boric acid of 100 mg/L + calcium nitrate of 200mg/L and the preferred processing parameters was the ultrasonic power of 150w,the working time of 5 seconds,the time interval of 5 seconds,the processing time of 8 seconds.Under the selection concentration of 200 mg/L of Kanamycin,3 T1 plants were obtained.[Conclusion] The experiment laid the basis for further genetic transformation of Chinese cabbage.
