白茅根及其复方汤对大鼠IgA肾病模型的干预作用 被引量:16

摘要 目的探讨白茅根及其复方汤对大鼠IgA肾病模型的影响。方法将50只大鼠分为对照组、模型组、白茅根组、茅根复方组与地塞米松组。采用牛血清白蛋白加葡萄球菌肠毒素B法建立IgA肾病大鼠模型,连续给药4周。检测大鼠尿红细胞计数(URBC)、24 h尿蛋白定量(UP)、血清肌酐(Scr)、尿素氮(BUN)、血清白介素-2(IL-2)、血小板活化因子(PAF)含量及肾组织病理变化;免疫组化及Western印迹法检测肾组织中转化生长因子-β1(TGF-β1)的含量及蛋白的表达。结果与模型组比较,各治疗组均可减少URBC、24 h UP、Scr、BUN、PAF,并降低TGF-β1蛋白表达,升高血清IL-2含量(P<0.01),减轻肾组织病理改变。结论白茅根及其复方汤对IgA肾病大鼠模型均可明显减少血尿、蛋白尿,减轻病理改变,改善肾功能。其机制可能是白茅根通过刺激机体分泌IL-2,抑制肾脏TGF-β1分泌与表达等而发挥作用。 Objective To investigate the effects of RhizomaImperatae(RI) and its compound decoction on the model of IgA nephropathy in rats. Methods 50 male rats were randomly divided into control group,model group,RI group,RI complex group,and dexame thasone group.Bovine serum albumin(BSA) plus staphylococcal enterotoxin B(SEB)method was used to establish IgA nephropathy rat model.Urine red blood cell(URBC) count,24hour urinary protein(UP),serum creatinine(Scr),blood urea nitrogen(BUN),serum interleukin-2(IL-2),platelet activating factor(PAF) content of rats were detected,and renal pathological changes were observed;Immunohistochemistry and western blot were renal tissue content of transforming growth factor(TGF-β1) and protein expression. Results Compared with the model group: URBC,24hUP,Scr,BUN,PAF and TGF-β1 expression were significantly decreased,serum IL-2 was significantly increased(P0.01) in the three treatment groups. Conclusion RI and its compound decoction can significantly reduce hematuria,proteinuria of the IgA nephropathy rats,reduce the pathological changes and improve renal function.The mechanism may be related to the elevation of IL-2 secretion of RI and inhibition of renal TGF-β1 secretion expression,etc.
