小鼠腹水型肝癌高低转移细胞株DARS2蛋白表达的差异 被引量:2

摘要 [目的]研究具有高、低转移潜能的小鼠腹水型肝癌细胞株Hca-F与Hca-P中线粒体的天门冬氨酰转移核糖核酸合成酶(DARS2)蛋白表达的差异。[方法]将Hca-F细胞与Hca-P细胞分别接种于615小鼠腹腔,抽取腹水,培养后收集细胞;用免疫印迹分析及免疫细胞化学方法检测Hca-F与Hca-P细胞的DARS2蛋白表达。[结果]DARS2在高转移细胞株Hca-F与低转移细胞株Hca-P表达量分别为(0.219±0.016)与(0.473±0.043),二者比较差异有显著性意义(P=0.01)。[结论]DARS2蛋白表达降低可能与小鼠肝癌转移相关。 [Objective] To research the expression difference of DARS2 in two murine hepatocarcinoma strains( Hca- F and Hca- P)with high and low metastatic potentials. [Methods~ The Hca- F cells and Hca -P cells were injected into abdominal cavity of 615 mice respectively, and then the ascitic cancer cells were drawn, cultured and collected. The pro- tein expressions of DARS2 in the two strains were detected by western blot and immunocytochemistry. [ Results ] The pro- tein expressions of DARS2 in Hca - F cells and Hca - P cells were O. 219 + O. 016 and O. 473 -+ 0. 043, respectively. The difference was significant (P =0.01 ). [ Conclusion] The decreasing expression of DARS2 protein may be related to me- tastasis potential of murine hepatocarcinoma.
