摘要 目的:探讨肿瘤坏死因子-α(Tumor necrosis factor-α,TNF-α)对3T3-L1脂肪细胞色素上皮衍生因子(Pigment epithelium-derived factor,PEDF)表达和分泌的影响。方法:体外培养和诱导分化3T3-L1前脂肪细胞。分别用不同浓度TNF-α(0.1、1、10、100 ng/ml)处理3T3-L1脂肪细胞24 h,并选取10 ng/ml的TNF-α干预细胞3、6、12、24 h,采用实时荧光定量PCR和ELISA技术检测3T3-L1脂肪细胞PEDF的mRNA表达和蛋白分泌情况。结果:①和对照组相比,不同浓度TNF-α对3T3-L1脂肪细胞PEDF的mRNA表达和分泌均有抑制作用,且浓度越大,其抑制效应越明显(P<0.05)。②10 ng/ml的TNF-α干预3T3-L1脂肪细胞后,随着时间的延长,PEDF的mRNA表达逐渐降低,第12 h达到最低点(P<0.05)。③10 ng/ml的TNF-α对其PEDF分泌的抑制率在第6 h为15%,在第24 h为33%(P<0.05)。结论:TNF-α抑制3T3-L1脂肪细胞PEDF的表达和分泌,其抑制效应呈剂量和时间依赖性。 Objective:To investigate the effect of tumor necrosis factor-α(TNF-α) intervention on the expression and secretion of pigment epithelium-derived factor(PEDF) in 3T3-L1 adipocytes.Methods:3T3-L1 preadipocytes were cultured and differentiated into the mature adipocyte.3T3-L1 adipocytes were incubated with TNF-α at different concentrations(0.1,1,10,100 ng/ml) for different periods of time(3,6,12,24 h).PEDF mRNA expression was detected by Real-time Quantitative PCR.Total protein of PEDF secreted by these adipocytes was evaluated by ELISA.Results:(1)Treatment of 3T3-L1 adipocytes with TNF-α at different concentrations decreased PEDF mRNA expression and secretion along with the elevated TNF-α concentration in the culture medium(P0.05).(2)The inhibitive effect of TNF-α(10 ng/ml) on PEDF mRNA expression gradually strengthened with the increase of treatment time,and reached the maximum at 12 h(P0.05).(3)Treated with TNF-α(10 ng/ml),the inhibition rate of their secretion of PEDF was 15% at 6 h,and 33% at 24 h(P0.05).Conclusion:TNF-α significantly decreases the expression and secretion of PEDF in 3T3-L1 adipocytes in a dose-and time-dependent manner.
