番茄转导外源DNA耐盐变异后代双向电泳分析 被引量:2

摘要 通过采用三氯乙酸(TCA)/丙酮法、酚提取法、DTT/丙酮法3种提取方法提取番茄叶片可溶性总蛋白,用150μg和250μg 2种加样量、浓度为12.5%和15%的聚丙烯酰胺凝胶二向电泳进行电泳,发现TCA/丙酮沉淀法提取番茄叶片可溶性总蛋白、加样量为250μg、12.5%聚丙烯酰胺凝胶二向电泳效果较好,从而建立了番茄叶片总蛋白双向电泳技术优化体系。用该体系对转导碱蓬总DNA的耐盐转化番茄叶片全蛋白的差异分析,鉴定出6个蛋白差异点,其中5个特异蛋白在耐盐转化番茄中表达,1个特异蛋白在未转化番茄中表达。
