摘要
以含油量不同的32份甘蓝型油菜自交纯合系为材料,用直接测序法筛查β-CT亚基的accD编码基因DNA序列的单核苷酸多态性,包括98%的编码区序列共1380 bp。在32份材料中只在材料18和20两个材料中发现了一个相同的SNP,SNP频率为1/1380,单倍型多态性指数Hd=0.121;核苷酸多态性π=0.00009,θ=0.00018。Tajima’s D值为不显著负值,Fu&Li’s检测,各个参数均为不显著正值,Z检验结果中(dN-dS)值也为不显著正值,说明accD基因没有偏离中性进化。系统发育树表明,accD基因在所选的32个油菜材料中是高度保守的,与拟南芥的序列相似度高达95.7%,说明accD基因在物种间也是高度保守的。暗示其在种子油脂合成过程中起着不可替代的作用,此外说明,仅用β-CT亚基编码基因的单核苷酸多态性还不能解释含油量复杂的表现型,还需在更大的群体资源中扩增accD编码基因和其它亚基编码基因进行单核苷酸多态性与含油量表型的相关性研究。
Taking 32 selfing homozygous lines of Brassica napus L. with different oil contents as test materials, the single nucleotide polymorphisms (SNPs) of β -CT subunit gene accD were screened by using direct sequencing method, it included 98% coding sequence, and its size was 1380 bp. Only one same SNP was found in accession 18 and accession 20 among all 32 accessions. Thus, SNP frequency was 1 SNP per 1380 bp, haplotype diversity index Hd = 0. 121, nucleotide diversity w = 0.00009 and 0 = 0.00018. The results of Tajima' s D tests in all regions of this gene expressed no significantly negative. All indexes of Fu & Li's tests and the value of dN - dS in Z - test expressed no significantly positive, it indicated that accD gene did not deviate from the Neutral Evolution. In ad- dition, the phylogenetic tree showed that the accD gene in 32 accessions was quite conservative during the evolution. The sequence similarity between this accD gene and that in Arabidopsis thaliana was up to 95.7%, indicating that the accD genes between species were also much conservative during the evolution. These results revealed that the accD gene played an irreplaceable role in the oil syn- thesis of the seed. Furthermore, the diversity of oil content could not be explained only by SNPs of accD gene. So the further studies on the SNPs and the oil content phenotype of rapeseed need to amplify accD gene and other subunit genes from much more accessions.
