摘要
目的探讨miR-126信号在脊髓源性神经干细胞定向分化为胆碱能神经元过程中的作用。方法采用神经干细胞球形成方法培养获得纯化的脊髓源性神经干细胞,添加诱导因子,诱导分化为胆碱能神经元,定量检测分化前后miR-126的表达变化。采用生物信息学方法,分析miR-126的靶基因和转录因子结合位点。结果 miR-126的表达在分化后的胆碱能神经元较未分化的神经干细胞明显增高。MiR-126作用的靶基因的作用包括蛋白结合、磷酸化修饰、神经发育、突触发育、细胞迁移、粘附等。miR-126含有3个潜在的转录因子结合位点。结论本文提示miR-126信号在神经干细胞向胆碱能神经元分化过程中具有调节作用。
Objective To explore the role of miR-126 signaling in the differentiation of neural stem cells(NSCs) into choline acetyltransferase-positive(ChAT) neurons.Methods The isolation of NSCs is performed through the classical neurosphere formation procedure.The NSCs were induced into ChAT-positive neurons with combined factors.The miR-126 expression was quantified using the TaqMan miR-126 Assay.The target gene and transcription factor binding sites were analyzed using the bioinformatic method.Results miR-126 was significantly elevated in ChAT-positive neurons differentiated from the NSCs.The functions of the target genes include protein binding,neural development,synapse development,cell migration and adhesion.Three transcription factor binding sites were predicted.Conclusion These results suggest miR-126 signaling can regulate the directed differentiation of NSCs into ChAT-positive neurons.
