摘要
本文对昆仑雪菊中总黄酮的提取进行研究,以得到较佳的提取工艺条件。在提取过程中通过单因素实验,分析了乙醇浓度、料液比、提取时间及回流温度等四个因素对提取率的影响并在此基础上建立正交实验,优化了总黄酮提取工艺条件。实验结果表明在最佳吸收波长510nm处,昆仑雪菊最佳提取工艺条件为:乙醇浓度60%,料液比1:60,回流温度80℃,提取时间4h,最佳黄酮含量为c=21.94%。以芦丁为对照品,用紫外可见分光光度法测定昆仑雪菊中总黄酮含量,方法简单,稳定,准确度较高。
During the extracting process, four main factors including concentration of extracting solution, solid-liquid rati- o ,refluxing time and temperature were studied by using single factor analysis method and based on the single-factor anal- ysis the L9 (34) orthogonal was designed and optimizes the best conditions extracting the total flavonoids from the C. tinctoria Flowers. The results showed that the best spectmphotometry at 510 nm, the optimal extracting industry condi- tion: methanol concentration 60%, solid-liquid ratio 1 : 60, refluxing time 4 h, refluxing temperature 80 ~C. Total fla- vonoids C =21.94%. so we can get the Conclusion that determine the content of flavonoids in C. tinctoria Flowers with ultraviolet-visual speetrophotometry by standard rutin is a practical, stable and simple method with higher accuracy.
