H_2O_2和ABA对干旱高温复合胁迫诱导的玉米叶片抗氧化防护基因表达的影响 被引量:7

摘要 为确定H2O2和脱落酸(ABA)对干旱高温复合胁迫诱导的苗期玉米(Zea mays L.)叶片抗氧化防护系统的影响,利用玉米ABA缺失突变体vp5及其野生型Vp5为试验材料,在不同胁迫下使用H2O2清除剂KI,ABA抑制剂钨酸钠(T)预处理后,测定了体内4个抗氧化防护基因SOD4,GR1,CAT1和cAPX的表达.结果显示:(1)与对照相比,干旱、高温和干旱高温复合胁迫均显著增加了SOD4,GR1,CAT1和cAPX等4个基因的表达,但除CAT1外,干旱高温复合诱导的增加量最高,其次是高温的;(2)KI和T预处理均明显抑制了干旱、高温和干旱高温复合胁迫诱导的4种抗氧化防护基因的表达;(3)与无ABA处理的vp5植株相比,100μmol ABA预处理均明显提高了干旱、高温和干旱高温复合胁迫诱导的4种抗氧化防护基因的表达水平.结果还表明,在干旱高温复合胁迫条件下,H2O2和ABA参与了抗氧化防护基因表达的调控,表明了H2O2和ABA是干旱高温复合胁迫诱导的玉米叶片抗氧化防护系统的重要调控因子. In order to determine the influences of H2O2 and ABA on antioxidant defense system induced by the combination of drought and heat stress in maize (Zea mays L. ) seedlings leaves, the ABA-defieient maize vp5 mutant and its wild-type Vp5 were used in these studies. After pretreatment with n2O2 scavenger KI,ABA inhibitor tungstate (T) , the expression of four antioxidant defense gene SOD4, GR1, CAT1 and cAPX were measured. The results were as follows: (1) compared with control,drought, heat and their combination stress all induced the gene expressions of SOD4, GR1, CAT1 and cAPX, but except CAT1, the increase induced by combined stress, then heat stress; (2) the pretreatment with KI and T significantly inhibited the genes expression of four enzymes induced by drought, heat and their combination stress; (3) compared with vp5 maize plant that without 100 μmol ABA pretreatment, ABA pretreatment significantly increased the four antioxidant defense gene expressions induced by drought, heat and their combination stress. These results suggeste that H2O2 and ABA participated in antioxidant defense genes expression in maize leaves exposed to the combination of drought and h sys Ke tem induce eat stress, which suggests that H2O2 and ABA might be important regulatory factors of d by the combination of drought and heat stress.
