摘要
目的 制备慢性阻塞性肺疾病(COPD)大鼠模型,研究黄芩苷对其防治作用及可能机制。方法成年雄性Wistar大鼠25只,分为空白组(F组),阳性对照组(C组),大、中、小剂量黄芩苷防治组(T1组、T2组、T3组),采用反复熏香烟加气管内两次灌注脂多糖的方法制备COPD大鼠模型,并于制备模型过程中进行干预。干预结束后处死大鼠,观察各组大鼠肺组织HE染色病理改变;测定平均肺泡数(MAN);肺组织干湿重比(RDW)及7干扰素(INF-γ)免疫组织化学阳性表达的灰度值。结果造模成功。各组大鼠肺组织RDW:F组15.36±0.43;C组10.86±0.51;T1组13.86±0.44;T2组13.16±0.42;T3组12.24±0.36。各组大鼠肺组织MAN:F组99.00±5.90;C组40.00±5.90;T1组65.00±5.20;T2组62.00±6.30;T1组58.00士4.50。各组大鼠肺组织INF-7免疫组织化学灰度值:F组108.71±7.85;c组60.94±2.21;T,组98.15±3.95;T2组91.17±2.45;T1组69.48土2.45。结论黄芩苷干预C()PD模型大鼠,可以改善大鼠肺部支气管炎、肺气肿的病理变化;提高其MAN、RDW;调节INF-γ在肺组织的表达,对大鼠COPD有一定的防治作用。黄芩苷防治COPD中的作用机制之一可能是调节INF-γ在肺组织的表达。
Objective To study prevention and treatment effects of Baicalin in model rats lung with chronic obstructive pulmonary disease (COPD) by made COPD model rats. Methods According to the principle of random, 25 Wistar rats(male) were divided into 5 groups, free group(group F), model contrast group (group C), maximum dose Baicalin, middling dose and minimum dose treatment groups (group T1, group T2, group T3 ). Model group was established by intratracheal instillation of lipopolysaccharide (LPS) twice and exposed to cigarette smoke. At the process of made model, gave Baicalin to treat. After treatment, killed the rats to research each group rats lung tissue HE dyeing pathological change and mean alveolar number (MAN) ,the ratio of dried lung weight to fresh lung weight (RDW), immunohistochemical stains were performed with INF-y to detect the expression. Results The COPD rats model were successfully made. The RDW of group F were 15.36±9 0.43, group C were 10.86±0.51,group TI were 13.86±0.44,group T2 were 13.16±0.42,group T3 were 12.24±0.36. The MAN of Group F were 99.00±5.90,group C were 40.00±5.90,group T1 were 65.00±5.20,group T2 were 62.00 ± 6.30, group T3 were 58.00±4.50, INF-y immunohistochemical stains were performed show group F were 108.71±7.85,group C were 60.94±2.21,group TI were 98.15±3.95, group T2 were 91.17±2.45,group T3 were 69.48±92.45. Conclusions Baicalin have some effects on COPD model rats. Maybe one of the theory is regulate INF-y expression in the lung.
