摘要
目的 探讨小干扰RNA(siRNA)对胃癌BGC823和MGC803细胞中Cullin1(Cul1)基因表达及细胞黏附力的影响.方法 体外化学合成Cul1 siRNA(si-Cul1)序列,同时合成Control siRNA(si-Ctrl)作为对照,在siLentFect Lipid Reagent介导下转染胃癌BGC823和MGC803细胞,Western blot检测Cul1蛋白及Src和FAK蛋白表达水平,用fibronectin包被96孔板行细胞黏附分析.结果 Cul1 干涉后胃癌BGC823和MGC803细胞株中Cul1、Src和FAK蛋白的表达均下降,同时胃癌细胞黏附于fibronectin包被板的能力降低.结论 Cul1 siRNA可以有效干涉胃癌BGC823和MGC803细胞中Cul1基因的表达,Cul1干涉后可能通过下调Src和FAK的表达水平进而降低胃癌细胞的黏附能力.
Objective To investigate the effects of small interfering RNA (siRNA) against Cullinl ( Cull ) gene on Cull expression and the adhesion of BGC823 and MGC803 gastric cancer cells. Methods Chemically synthesized small interfering RNA targeting Cull ( si - Cull ) was transfeeted into human gastric cancer cell lines BGC823 and MGC803 by siLentFect Lipid Reagent. Control siRNA ( si - Ctrl ) was used as a control. The expression levels of Cull, Src, and FAK proteins were detected by Western blot. Cell attachment was detected by fibronectin - coated ninety - six - well plates. Results The expression levels of Cull, Src, and FAK proteins were decreased in BGC823 and MGC803 gastric cancer cells after Cull knockdown, and cell attachment ability of gastric cancer cells was also decreased in fibronectin - coated plates. Conclusion Cull siRNA can effectively interfere the expression of Cull gene in both BGC823 and MGC803 gastric cancer cells. Cell attachment ability of gastric cancer cells may be decreased after Cull knockdown by down- regulating Src and FAK expression.
