摘要
目的探讨microRNA-224(miR-224)在肝细胞癌(hepatocellular carcinoma,HCC)患者癌组织与血浆中表达的相关性。方法采用qRT-PCR检测20例HCC患者的癌组织、癌旁组织、术前血浆及20例健康人血浆的miR-224表达量,比较癌组织与癌旁组织、HCC患者与健康人血浆miR-224的表达差异,分析miR-224在HCC血浆与癌组织中表达的相关性。结果①miR-224在HCC癌组织、癌旁组织、血浆及健康人血浆中的平均表达量分别为(0.14±0.04)、(0.02±0.01)、(1.15±0.26)和(0.11±0.03)。癌组织miR-224表达量高于癌旁组织(P<0.01),HCC患者血浆miR-224表达量高于健康人血浆(P<0.01);②miR-224在HCC血浆及相应癌组织中的相对表达量呈正相关(P<0.001)。结论 HCC血浆miR-224表达量高于健康人群,并与肝癌组织表达量呈正相关,血浆miR-224有可能成为一种新的血清学指标用于肝癌诊断。
[Objective] To investigate the correlation of microRNA-224 (miR-224) expression between plasma and tumorous tissue in HCC patients. [Methods] The expression of miR-224 in HCC tumorous tissues, adjacent nontumorous tissues and plasma was detected by qRT-PCR in 20 cases of HCC patients, and the expression of miR-224 was also detected in 20 cases of healthy human plasma, the difference of miR-224 expression between tumorous tissues and adjacent nontumorous tissues was compared, and the difference of miR-224 expression between HCC plasma and healthy human plasma was also compared, then the correlation of miR-224 expression between plasma and tumorous tissue in HCC patients was analyzed. [Results ] The average of miR-224 expression in HCC tumorous tissues, adjacent nontumorous tissues, plasma and healthy human plasma were (0.14±0.04), (0.02±0.01), (1.15±0.26), (0.11±0.03), (P〈0.01), respectively. The expression of miR-224 in tumorous tissues was significantly higher than that in adjacent nontumorous tissues (P 〈0.01), the expression of miR-224 in HCC plasma was significantly higher than that in healthy human plasma (P〈0.01). There was a positive correlation of miR-224 expression between plasma and tumorous tissue in HCC patients (P〈0.001). [Conclusions] The expression of miR-224 in HCC plasma is significantly higher than that in healthy human plasma, the expression of miR-224 in HCC plasma is positively correlated with that in HCC tumorous tissue. The plasma miR-224 may be a new serum biomarker for diagnosis of HCC.
