摘要
目的分析不同条件制备的血府逐瘀汤含药血清促血管内皮细胞增殖能力的差异。方法采用MTT法检测HUVE-12细胞活性来反应其增殖能力;通过不同时间点制备血府逐瘀汤含药血清,分别采用蛋白沉淀、灭活等方式处理含药血清,比较其促HUVE-12增殖能力的差异;采用ELISA、分光光度法和HPLC检测各组含药血清的细胞因子水平和成分,最后选择增殖作用最强的含药血清考察其在培养体系中体积量对HUVE-12增殖能力的影响。结果在血府逐瘀汤含药血清对HUVE-12细胞增殖作用比较时,灌胃给药7 d制备的血府逐瘀汤含药血清作用优于其它时间点;未灭活作用优于灭活处理;未蛋白沉淀处理作用明显优于蛋白沉淀处理;含药血清加入量占细胞培养体系5%~10%为宜。结论在研究血府逐瘀汤促HUVE-12增殖作用机制时,可采用灌胃给药7天制备其含药血清,不作蛋白沉淀、灭活等方式处理。
Objective To analyze the proliferative activity of vascular endothelial cells(VEC) induced by serum containing Xuefu Zhuyu decotion which were prepared in different conditions.Methods The activity of HUVE-12 cells was detected by MTT assay.Serum containing Xuefu Zhuyu decotion was prepared in different time,and treated by protein precipitation or inactivation,then their effects of promoting VEC proliferation were compared.The levels and ingredients of cytokines of serum containing Xuefu Zhuyu decotion were evaluated by ELISA,spectrophotometry and HPLC.Finally,the serum containing which showed the strongest proliferation effect was chosed,and the effects of different amount of the serum containing on proliferative capability of HUVE-12 cells were observed.Results After administration of gastric infusion was executed for 7 days,the effect of serum containing Xuefu Zhuyu decotion was better than that in other time.The effect of serum containing which was not treated by inactivation was better than that treated by inactivation.The effect of serum containing which was not treated by protein precipitation was better than that treated by protein precipitation.The amount of serum containing accounted for 5%-10% of the total amount of cell culture system.Conclusion Serum containing Xuefu Zhuyu decotion is prepared after administration of gastric infusion was executed for 7 days,and not treated by protein precipitation or inactivation,which is the best condition for studying the mechanism of promoting VEC proliferation of serum containing Xuefu Zhuyu decotion.
