摘要
目的观察高脂饮食对大鼠肝脏PGC-1α、TRB3、Akt蛋白水平及血浆脂联素的影响,探讨其引起胰岛素抵抗的机制。方法将24只雄性Wistar大鼠按照体重随机分为两组,分别给予普通饮食和高脂饮食,22周后处死大鼠,测量体重、体脂重量,取血检测血脂、血糖、胰岛素和脂联素;取肝脏检测PGC-1α、TRB3、Akt蛋白水平。结果与对照组相比,高脂饮食组大鼠空腹血糖、胰岛素和胰岛素抵抗指数分别增加27.1%(P>0.05)、27.3%(P<0.05)和68.8%(P<0.01),血浆中脂联素含量降低,空腹血清TC、HDL-C、LDL-C及TG水平和正常对照组大鼠无显著性差异;高脂饮食组大鼠肝脏PGC-1α蛋白水平上升(P<0.05),TRB3蛋白水平上升(P<0.05),Akt蛋白水平下降(P<0.05)。结论高脂饮食可诱导大鼠产生肥胖和胰岛素抵抗,可能和PGC-1α/TRB3/Akt信号通路有关。
Objective To observe the effects of high-fat diet on plasma adiponectin and PGC-1 alpha,TRB3,Akt protein levels in rat liver,and discuss the mechanism on insulin resistance. Methods Twenty-four male Wistar rats according to weight were randomly divided into two groups,separately given ordinary diet and high-fat diet for 22 weeks;Then measured body weight,body fat weight,collected the blood and tested blood fat and blood glucose,insulin and adiponectin;Collected the liver tissues to detect PGC-1 alpha, TRB3 and Akt protein levels. Results Comparing with the control group,fasting blood glucose,fasting insulin and HOMA-IR index of high-fat diet group were increased by 27.1%(P〉0.05),27.3%(P〈0.05) and 68.8%(P〈0.01),plasma adiponectin level was reduced; Fasting serum TC,HDL-C,LDL-C and TG levels had no significant difference with the control group. In high-fat diet group,rat liver PGC-1α protein level was increased(P〈0.05),TRB3 protein level was increased(P〈0.05),Akt protein level was reduced(P〈0.05). Conclusion High fat diet can induce rat to be obesity and insulin resistance,and PGC-1 α/TRB3/Akt signaling pathway might be involved.
