摘要
目的研究葡萄籽原花青素(GSP)对DOCA-盐型高血压小鼠心室重构保护作用并初步探讨其机制。方法皮下注射去氧皮质酮(DOCA)同时予高盐饮水形成DOCA-盐型高血压小鼠模型。实验分为对照组、模型组和GSP 130 mg.kg-1组。计算小鼠心脏重量指数以及肾脏重量指数,HE染色和V-G染色观察心肌病理形态学改变,比色法测定血清中超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量以及心肌羟脯氨酸(Hyp)含量。结果与空白对照组相比,DOCA-盐型高血压模型组小鼠心重指数、肾重指数、心肌细胞横截面积(CSA)、心肌胶原纤维明显升高,血清MDA含量升高,SOD活性降低,心肌组织羟脯氨酸含量增加。与模型组相比,GSP 130 mg.kg-1组小鼠心重指数、肾重指数、CSA、心肌胶原沉积明显降低,血清MDA含量降低,SOD活性增加,心肌组织Hyp含量减少。结论 GSP能明显改善DOCA-高盐高血压小鼠心室重构,其机制可能与抗氧化应激有关。
Aim To investigate the protective effects and mechanism of oligomeric grape seed proanthocyanidins(GSP) on ventricular remodeling of DOCA-salt hypertensive mice.Methods DOCA was given subcutaneously(150 mg·kg^- 1 sc,twice per week for 4 weeks) and 1% saline;0.2 % potassium chloride was added to their tap water for drinking to induce cardiac remodeling in mice.Mice were divided into 3 groups:control group,model group and GSP 130 mg·kg^-1 group.The heart weight index and kidney weight index were expressed as heart weight/body weight(HW/BW)and kidney weight/body weight(KW/BW).The histological changes were investigated by HE and Van Gieson staining.Colorimetric analysis was used to assay SOD activity,MDA content in serum and hydroxyproline(Hyp) content in cardiac tissue.Results Compared with control group,the heart weight index,kidney weight index,cardiomyoctytes cross section area(CSA),cardiac collagen deposition and cardiac Hyp content as well as serum MDA markedly increased,and serum SOD activity was decreased significantly.Administration of GSPs markedly alleviated the elevation of HW/BW,KW/BW ratio and CSA of cardiomyocytes,decreased collagen deposition in heart.Meanwhile,treatment with GSPs significantly ameliorated oxidative stress via improving SOD activities and decreasing MDA formation.Conclusion GSP has protective effects against cardiac remodeling of DOCA-salt induced hypertensive mice,which may be related to the reduction of oxidative stress.
