滋阴活血解毒方对局灶性脑缺血大鼠凝血酶原mRNA和蛋白酶激活受体-1 mRNA含量的影响

摘要 目的观察滋阴活血解毒方对局灶性脑缺血模型大鼠脑组织凝血酶原mRNA和蛋白酶激活受体(PAR-1)mRNA表达的影响,探讨其脑保护机制。方法将24只大鼠随机分为假手术组、模型组、滋阴活血解毒方组、阿加曲班组,每组分1、3 d两个时间点。滋阴活血解毒方组灌服中药水煎剂,每日剂量32.4 g/kg;阿加曲班组为腹腔注射给药,第1、2日每日剂量为11.16 mg/kg,第3日剂量为3.72 mg/kg。假手术组和模型组均灌服等容量生理盐水,每日剂量为2 mL/100 g。均分2次给药。线栓法制备大脑中动脉闭塞模型。采用反转录-聚合酶链反应(RT-PCR)检测脑组织中凝血酶原mRNA及PAR-1mRNA表达水平。结果与假手术组比较,模型组脑组织中凝血酶原mRNA、PAR-1 mRNA表达显著升高,差异具有统计学意义(P<0.01),滋阴活血解毒方组和阿加曲班组能明显下调凝血酶原mRNA、PAR-1 mRNA表达,差异具有统计学意义(P<0.01)。结论滋阴活血解毒方能抑制脑组织中凝血酶原mRNA和PAR-1 mRNA的表达,减轻凝血酶的毒性反应,这可能是其发挥脑保护作用的机制之一。 Objective To observe the effect of Ziyin Huoxue Jiedu decoction on the expression of prothrombim mRNA and protease-activated receptor-1 (PAR-1) mRNA of focal cerebral infarction rats. Methods Twenty-four rats were randomly divided into 4 groups: the sham-operated group, ischemic group, Ziyin Huoxue Jiedu decoction group, Argatroban group. Each group was divided into two time phase point of 1, 3 d. Ziyin Huoxue Jiedu decoction group was fed with Chinese medicine decoction with the daily dose of 32.4 g/kg. Argatroban group was given the intraperitoneal injection, with the daily dose of 1, 2 d 11.6 mg/kg, day 3 dose of 3.72 mg/kg. The sham-operated group and ischemic group were fed with saline with the daily dose of 2 mL/100 g. Daily dose divided equally into 2 times. The model was reproduced by occluded middle cerebral artery. The expression of prothrombin mRNA and PAR-1 mRNA of brain tissue were detected by reverse transcription-polymerase chain reaction (RT-PCR). Results The expression of prothrombin mRNA and PAR-1 mRNA of brain tissue in model group was significantly higher than that in the sham-operated group (P〈 0.01), and the expression of prothrombin mRNA and PAR-lmRNA of brain tissue in Ziyin Huoxue Jiedu decoction group and Argatroban group was significantly lower than that in the model group (P〈0.01). Conclusion Ziyin Huoxue Jiedu decoction is effective to restrain the expression of prothrombin mRNA and PAR-1 mRNA of brain tissue, reducing thrombin toxic injury, which may be one of the mechanisms of cerebral protective effects of Ziyin Huoxue Jiedu decoction.
