Effects of the estrogen replacement therapy on the NGFR in the basal forebrain and on the Cognitive function of the ovariectomized rats

摘要 目的 探讨雌激素替代对切卵巢大鼠基底前脑胆碱能神经元神经生长因子表达的影响和对学习记忆的影响。方法 50 只雌性切卵巢大鼠分成给雌激素组和对照组,分别用免疫组化方法测定给药后1 周、2 周、4 周时基底前脑各部位神经生长因子受体(NGFR)阳性神经元数,用MORRIS迷宫观察给药4 周时大鼠逃避潜伏期和搜索策略,并进行两组间比较。结果 给雌激素组与对照组的NGFR 阳性神经元数相比,内侧隔核(MS)和Meynert 基核(NBM) 部位分别在2 周和4 周开始有显著性降低,而斜角带水平肢(HDB)在4 周内无显著性变化。给药4 周后给雌激素组与对照组相比,逃避潜伏期延长有显著性而空间搜索策略差异无显著性。结论 ①雌激素替代对基底前脑胆碱能神经元的神经生长因子受体的表达有时间性和部位选择性作用。②给雌激素4 周时,大鼠的学习获取能力减退,但空间定位、学习巩固和记忆储存能力尚无明显变化。 Objective To investigate the effects of estrogen replacement therapy(ERT)on the cholinergic system nerve growth factor related system in the basal forebrain and on the cognitive function.Methods Fifty female ovariectomized rats were divided into two groups(estrogen replaced therapy group and no estrogen treated control group).After 1 week、2 week and 4 week therapy,the rats were executed,and the NGFR immunoreactive cells in the basal forebrain were detected by immunocytochemical method.Morris mazes were used to evaluate the memory and learning ability of the ovariectomized rats after 4 weeks of ERT.Results The number of NGFR immunoreactive cells of ERT rats compared with that of control rats was as follows:In the MS,there was a significant decrease after 2 weeks.In the NBM,there was a significant decrease after 4 weeks.In the HDB,there wasn't signnificant change within 4 weeks.In the place navigation test of Morris mazes,escape latencies of the ERT rats were significantly increase the first 4 blocks compared with those of the controls(30.4±7.5svs 21.3±5.3s,P<0.01);in the latter 5 blocks escape latency had no significant difference between the two groups(13.2±7.7vs 9.8±3.1,P>0.05).In the spatial probe test of Morris mazes,there was no significant difference between ERT group and control group in the percentage of the swimming distance in the every quadrant part over the total swimming area.Conclusion ①ERT produces regionally selective effects on the NGFR expression of the basal forebrain cholinergic neurons.②Short term(1 month)ERT makes the ovariectomized rats more difficult to obtain new information,but exerts no influence on the other aspects of the memory and learning.
