益肾降浊冲剂干预肾小球硬化的实验研究 被引量:5

Protective effect and mechanism of Yishenjiangzhuo Granules on renal glomerulosclerosis

摘要 目的:探讨益肾降浊冲剂对阿霉素肾病大鼠肾小球硬化的干预作用与机制。方法:SD大鼠随机分为对照组、模型组、他汀组、益肾降浊冲剂(低、中、高剂量)组,每组6只。采用单侧肾切除加阿霉素重复注射建立肾小球硬化模型。用药8周后观察各组大鼠24h尿蛋白定量、血生化、氧化低密度脂蛋白(ox-LDL)的变化,PAS、Masson染色评估肾小球硬化程度,免疫组化观察肾小球氧化低密度脂蛋白受体(LOX-1)的表达,Western Blot检测Ⅰ型胶原(ColⅠ)、纤连蛋白(FN)的表达。结果:与对照组比较,模型组大鼠24h尿蛋白量、尿素氮(BUN)、肌酐(Scr)、胆固醇(Cho)、低密度脂蛋白胆固醇(LDL-C)、ox-LDL、肾小球硬化评分、系膜肾小球系膜基质比(M/G)均明显升高,肾小球LOX-1、ColⅠ、FN表达显著增加(P<0.01)。与模型组比较,益肾降浊冲剂组呈剂量依赖性降低24h尿蛋白量、Bun、Scr、Cho、LDL-C、ox-LDL水平以及肾小球硬化评分、M/G(P<0.01),下调LOX-1、ColⅠ、FN的表达(P<0.01)。结论:益肾降浊冲剂组能通过纠正脂代谢紊乱、抑制LOX-1的表达、减轻细胞外基质的沉积、改善肾小球硬化。 Objective: To investigate the protective effect and its possible mechanism of Yishenjiangzhuo Granules on renal glomerulosclerosis in the adriamycin(ADR)-induced nephropathy rats.Methods: The Sprague-Dawley(SD) rats were randomly divided into six groups: control group,model group,atorvastatin group,high,medium and low doses of Yishenjiangzhuo Granules group.Every group had six rats.The model was established by adriamycin injection and unilateral nephropathy.After treated for 8 weeks,PAS and Masson staining was used to assess glomerulosclerosis.The 24-hour urinary protein excretion and renal function were examined.Immunohistochemistry and Western Blot were used to messure the expression of LOX-1,ColⅠand FN.Results: Compared with control group,the 24-hour urinary protein excretion,BUN,Scr,Cho,LDL-C,ox-LDL was significantly increased in model group,as well as more severe glomerulosclerosis(P0.01).The expression of LOX-1,ColⅠand FN were significantly enhanced(P0.01).Yishenjiangzhuo Granules groups significantly decreased the above indexes in a dose dependent manner(P0.01).Conclusion: Yishenjiangzhuo Granules can alleviate the dyslipidemia,down-regulate the expression of LOX-1,ColⅠand FN,and then inhibit glomerulosclerosis.
