Expression and Distribution of LRRN3 Membrance Protein in Human Embryo Dorsal Root Ganglion(DRG)

摘要 目的:探讨神经系统富亮氨酸重复超家族成员LRRN3膜蛋白在人胚脊神经节的表达和分布。方法:从人胚脊神经节分离mRNA和蛋白质,用RT-PCR、Western印记杂交法和免疫组织化学方法检测LRRN3膜蛋白表达。结果:LRRN3膜蛋白C端序列RT-PCR扩增产物cDNA在各胎龄脊神经节均有表达,长度约500bp;Western印记杂交结果显示LRRN3膜蛋白存在,分子质量约78 kD;免疫组织化学和免疫荧光组织化学结果表明LRRN3阳性表达细胞均为脊神经节感觉神经细胞,部分神经细胞弱阳性表达,部分未表达。结论:LRRN3膜蛋白在脊神经节神经元的表达,推断与脊神经节感觉神经元的发育、形态构建及损伤后修复有密切的关系。 Objective: To explore the expression and localization ofmembrance protein LRRN3 of dorsal root ganglion (DRG) in human embryo at the mRNA and protein levels. Methods: mRNA and protein of membrance protein LRRN3 were separated from DRG, and RT-PCR, Western Blotting, immunocytochemistry staining and Immunofluorescence were used to detect expression of membrance protein LRRN3. Results: RT-PCR production eDNA of LRRN3 C-terminal domain protein were detected in DRG at each gestational age, and the length is about 500 bp; Western blot analysis showed membrance protein LRRN3 were expressed and the relative molecular mass is about 78 kD; immunocytochemistry staining and Immunofluorescence detection showed all membrance protein LRRN3 positively expressed cells were sensory neurons in DRG, some weak-positively expressed and some not expressed. Conclusion: The expression of LRRN3 in DRG suggests that LRRN3 is closely associated with embryonic development, morphogenesis and repair following injury of sensory neurons in Dorsal root ganglion.
