逆电针“足三里”穴对运动大鼠下丘脑、纹状体DA、5-HT含量/比值的影响 被引量:5

Effect of pre-electroacupuncture at "Zusanli" (ST 36) on DA and 5-HT contents and their ratio in hypothalamus and striatum in exercise rats

摘要 目的:观察逆电针"足三里"穴对急性跑台运动大鼠的抗疲劳效应及其中枢机制。方法:将清洁级雄性SD大鼠50只随机分为安静组(n=10)、模型组(n=20)和逆电针组(n=20),后两组又分为运动后即刻、运动后3h组(各10只)。后两组大鼠完成适应性跑台运动后,接受急性跑台运动;逆电针组大鼠于跑台运动前先介入电针治疗(参数为双侧"足三里"穴、连续波、频率2Hz、强度2mA、时间30min、1次/d、连续6d)。分别于运动后即刻和运动后3h处死大鼠,检测血浆乳酸水平、下丘脑和纹状体多巴胺(DA)、5-羟色按(5-HT)含量变化,并计算DA/5-HT比值。结果:与安静组相比,模型组大鼠血浆乳酸和下丘脑5-HT水平呈升高趋势,下丘脑DA含量则显著升高(P<0.01);而纹状体DA和5-HT含量于运动后3h时显著降低(均P<0.01)。运动后即刻时,与同期模型组相比,逆电针组大鼠下丘脑DA和5-HT含量显著升高(均P<0.01),而纹状体则明显下降(均P<0.01);运动后3h时,与同期模型组相比,逆电针组大鼠血浆乳酸水平、下丘脑5-HT含量均显著降低(P<0.05,P<0.01),而下丘脑DA/5-HT比值则明显升高(P<0.01)。结论:逆电针"足三里"穴可促进急性跑台运动大鼠疲劳恢复,可能是通过降低跑台运动大鼠血浆乳酸堆积,提高下丘脑DA/5-HT比值发挥作用。 Objective To observe anti fatigue effect and mechanisms of pre--electroacupuncture (EA) at "Zusanli" (ST 36) in rats undergoing acute treadmill running. Methods Fifty male SI) rats were randonfly divided into three groups: a quiet group (group Q, n= 10), a model group (group M, n=20) and an EA preconditioning group (group EAP, n=20). After adaptation for undergoing treadmill running, all the rats in group M and group EAP were trained on acute treadmill running. Besides, EA with continuous waves, 2 Hz in frequency and 2 mA in intensity was applied at bilateral "Zusanli" (ST 36) for 30 nfin, which was applied once daily for contirmous 6 days before treadmill running for the rats in Group EAP. Plasma laclate contents were measured immediately and 3 hours after treadmill running, respectively. Changes of dopamine(DA) and serotonin(5-HT) contents obtained immediately and 3 hours after treadmill running, respectively, in hypotimlamus and stfiatum, were. detected and compared, and DA/5-HT ratios were calculated. Results Compared with group Q, the levels of blood lactate and hypot[mlamic 5-HT tented to increase in rats of group M, and the contents of hypothalamic DA increased significantly (P〈 0.01) .while the contents of stfiatal DA and 5-HT in group M decreased significantly (both P〈0. 01) at 3 h after treadmill run-- ning. Immnediately after treadmill running, the contents of DA and 5-HT increased significantly in hypottmleanus (both P〈0. 01), but decreased significantly in striatum (both P〈0. 01) in group EAP, compared with those in group M. Morc(wer, EA pretreatment markedly decreased the levels of blood lactate (P-〈0. 05) and hypothalanic 5 HT (P〈0. 01), and obviously elevated the ratio of DA/5-HT in the hypothalamus (P〈0. 01) at 3 h after treadmill running. Conclusion Preventive EA at "Zusanli" (ST 36) can accelerate recovery from fatigue, which nmy be related to its reducing accumttlation of blood lactate, elevating DA
