摘要
目的:筛选分离纯化骨碎补中柚皮苷的最佳树脂型号及最佳工艺。方法:通过静态、动态吸附和解吸相结合的方法,以柚皮苷的吸附率、解吸率为评价指标综合评判确定树脂型号;通过考察上柱药材量、上样流速考察、洗脱乙醇浓度、洗脱溶剂用量,最终确定了纯化工艺条件。结果:HPD-100型大孔吸附树脂分离纯化骨碎补中柚皮苷效果最好,其最佳工艺条件为:以药材∶大孔吸附树脂为1∶1(质量比)上样,上样流速为3 BV.h-1,以8倍柱体积水洗,继以14倍柱体积50%乙醇洗脱。结论:采用HPD-100型大孔吸附树脂分离纯化骨碎补中柚皮苷效果良好。
OBJECTIVE To screen the best resins model and the best craft for isolation and purification of naringin from Drynaria. METHODS Through the combination of static and dynamic adsorption and desorption methods, taking naringin adsorption rate, desorption rate as the evaluating index for comprehensive evaluation and determination of resin models; by exam ining the amount of herbs on the column, studying on the sample flow, ethanol elution and elution solvent to determine ultimately the purification procedure. RESULTS HPD-100 type macroporous resin was the best for purification of naringin from Drynaria, the best conditions were: macroporous resin 1:1 (mass ratio) on the sample, sample flow rate 3 BV.h-1 , washed with 8 times column volumes of water, followed by 14 times column volume of 50% ethanol. CONCLUSION Using HPD-100type macroporous resin for purification of naringin from Drynaria can get good result.
