HPLC法测定江西不同产地蔓荆子中蔓荆子黄素的含量 被引量:7

摘要 目的:建立测定江西不同产地蔓荆子中蔓荆子黄素含量的方法。方法:采用高效液相色谱法。色谱柱为Alltima HP C18(250mm×4.6mm,5μm),流动相为乙腈-50mmol·L-1磷酸二氢钾溶液(磷酸调pH值为3.0)=50∶50,流速为1.0mL·min-1,检测波长为254nm,柱温为室温,进样量为10μL。结果:蔓荆子黄素的进样量在0.2~1.2μg范围内与峰面积积分值呈良好的线性关系(r=0.9994);平均加样回收率为99.96%,RSD=1.42%(n=9)。江西不同产地蔓荆子中蔓荆子黄素含量差异较大。结论:本方法简便、快速、线性关系良好,可用于蔓荆子中蔓荆子黄素的含量测定。 OBJECTIVE: To establish a method for the content determination of vitexicarpin in Vitex trifolia from different pro- ducing areas of Jiangxi province. METHODS: HPLC method was adopted. The determination was performed on Alltima HP C18(250 mm×4.6 mm, 5 μm) column with the mobile phase consisted of acetonitrile-50 mmol.L-1 potassium dihydrogen phosphate solution (pH value adjusted to 3.0 with phosphoric acid, 50:50), at the flow rate of 1.0 mL.min-1. The detection wavelength was set at 254 urn, and the column temperature was room temperature. The injection volume was 10 μL. RESULTS: The linear range was 0.2~ 1.2μg(r= 0.999 4) with an average recovery of 99.96 % (RSD= 1.42 %, n =9).Vitexicarpin contents of E trifolia from different areas differed greatly. CONCLUSION: The method is simplp and rapid with good linear relationsgip, which can be used for the content determination of vitexicarpin in V. trifolia.
