摘要
目的:观察参附注射液对大鼠心室肌细胞L型钙通道的作用。方法:采用急性酶解法获得单个心肌细胞,全细胞膜片钳技术记录钙电流。结果:参附注射液浓度依赖性阻断L型钙通道电流,其IC50为原液2.5%稀释。参附注射液(0.3%,3%)能够上移I-U曲线,但不改变峰电位和反转电位;3%参附注射液对L型钙通道激活曲线失活曲线无明显影响,但可使通道电流从失活中恢复的时间明显延长,时间常数由给药前的82ms增至给药后的139ms。结论:参附注射液对心肌细胞L型钙通道电流有抑制作用。
Objective: To study the effects of Shenfu injection(SF)on L-type calcium currents of ventricular myocytes in rats.Methods: The whole cell patch clamp technique was used to measure the L-type calcium currents in single myocytes dissociated by acute enzymatic dissociation method.Results: SF could block the L-type calcium currents in a concentration-dependent manner,IC50=2.5%;SF(0.3%,3%)made the I-V curves upward and did not change the reverse potential.SF in the concentration of 3% had no effect on the steady-state activation curves and inactivation curves,but the recovery duration from inactive state of L-type calcium currents prolonged significantly and the time constantly increased from 82ms to 139ms.Conclusion: SF exerts blocking effect on L-type calcium currents.
