摘要
目的:研究柴胡总皂苷粗提物单次给药致小鼠肝毒性的时毒、量毒关系。方法:小鼠按不同时间点或不同剂量分组,观察小鼠死亡情况和毒性反应,检查血清谷丙转氨酶(ALT)、谷草转氨酶(AST)活性,计算肝、脾、胸腺脏器指数,光学显微镜下观察肝组织形态变化。结果:柴胡总皂苷粗提物31.25 g/kg剂量组对肝组织产生明显损伤,且随着剂量增大,ALT、AST升高显著,均在给药后4 h升高并达到高峰,48 h可恢复近正常值。光学显微镜下观察,柴胡总皂苷粗提物组小鼠肝组织出现不同程度的肝细胞水肿和脂肪变性,其中高剂量组甚至出现片状坏死、小叶结构不清。结论:小鼠单次灌服一定剂量柴胡总皂苷粗提物可造成急性肝损伤甚至死亡,并呈现一定的时毒、量毒关系。
Objective: To study on the time-toxicity and dose-toxicity relationships caused by single dose total Bupleurum saponin extracts to mice.Methods: Mice are grouped according to different time or dose points,to observe the death condition and toxicity of mice,detect ALT 、AST in serum,calculate liver,spleen,thymus index,and observe the morphological changes of hepatic tissue under light microscope.Results: ALT,AST in serum are peaked after 4 hours' administration,and restored near normality after 48 hours.Compared with the normal group,31.25 g/kg dose group of total Bupleurum saponin extracts produced significant hepatic injury,and ALT,AST in serum increased significantly with the dose increasing.Under light microscope,the hepatic tissue of different dose mice all had different degree's edema and fatty degeneration to hepatic cells,and the high dose group even appeared to be necrosis and lobular structure unclear.Conclusion: The total Bupleurum saponins extracts gavaged certain dose one time to mice could cause acute hepa-injury and even death,and there were certain time-toxicity and dose-toxicity relationships.
