摘要
目的:探讨湖北海棠总黄酮对去势大鼠骨质疏松的影响。方法:取三月龄SD雌性大鼠经去势手术或假手术后,喂以去大豆特殊饲料7周以诱导骨丢失。将去势大鼠分为模型组,湖北海棠总黄酮组(23.3、46.6、93.3mg/kg),己烯雌酚组,连续给药12周。检测大鼠血清中骨钙素,雌二醇的含量,尿液中脱氧吡啶啉的含量,血清和尿液中Ca、P含量,观察大鼠腰椎骨末端骨小梁的变化情况并计算骨小梁面积百分比。结果:湖北海棠总黄酮降低了大鼠血清中骨钙素以及尿液中脱氧吡啶啉的含量,与模型组相比,46.6mg/kg和93.3mg/kg剂量组差异有显著性;湖北海棠总黄酮提高了大鼠血清中雌二醇含量,但与模型组相比,无统计学意义;湖北海棠总黄酮提高了大鼠血清钙含量,降低大鼠尿液中钙、磷的含量,93.3mg/kg组作用最显著;骨小梁的形态学观察及数据分析显示,与模型组相比,湖北海棠总黄酮可以提高骨小梁的厚度和面积,其46.6 mg/kg和93.3mg/kg明显地提高了骨小梁面积百分比。结论:湖北海棠总黄酮可以降低去势大鼠骨转换率和骨吸收程度,对绝经后骨质疏松具有防治作用,其作用机制可能与湖北海棠总黄酮中根皮苷等黄酮成分具有雌激素类似结构,能与雌激素受体结合,发挥雌激素样作用有关。
Objective: The purpose of this study was to examine the osteoporotic effects of Malus hupehensis(Pamp.) Rehd.(MH) flavonoids in estrogen-deficient ovariectomized rats.Methods: Three-month-old female Sprague-Dawley rats were either sham-operated or ovariectomized(OVX) and fed on special diet without soybeen for 7 weeks to induce bone loss.The OVX rats were further divided into five subgroups respectively,with vehicle(OVX),MH(23.3,46.6,93.3mg/kg),diethylstilbestrol for 12 weeks.The concentration of osteocalcin,calcium,phosphorus,estradiol in serum,calcium,phosphorus,deoxypyridinoline in urine,the variation of proximal end of the vertebrae and the area percentage of the trabecular bone of the vertebra were determined.Results: Compared to model group,administration of MH decreased serum osteocalcin and urine deoxypyridinoline.And significant differences were observed in MH treated rats(46.6 mg/kg and 93.3mg/kg).The MH groups increased serum E2 content and no significant difference was observed.Serum calcium level was increased in MH treated rats and significant differences were observed in the group of MH 46.6 mg/kg and 93.3mg/kg.Urine calcium and phosphorus levels among the MH groups were lower than that of OVX group and the group of MH 93.3mg/kg showed significant difference.The MH groups increased the thickness and the area percentage of the vertebra of the trabecular bone of ovariectomized rats,the effect of 46.6 mg/kg and 93.3mg/kg were significantly different compared with OVX group.Conclusion: MH has a definite protective effect on bone loss in ovariectomized rats by inhibiting bone turnover and bone resorption.The possible mechanism may be that Phloridzin,the main compound in MH,could produce estrogen-like effects because of their structural similarity to estradiol and combination with estrogen receptors(ERs).
