摘要 目的研究甘肃地区汉族人群AMPKα2基因多态性与T2DM、胰岛素抵抗及血清脂联素、抵抗素水平的关系。方法选取T2DM患者188例和非糖尿病正常人群179例,用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)法,检测AMPK-rs2796516G/A基因型。用酶联免疫法(ELISA)测定血清脂联素、抵抗素浓度,以及测定血糖、血脂、胰岛素(INS)。结果 T2DM和正常人群中均存在AMPK-rs2796516位点G/A基因突变,二组间基因型构成比及等位基因频率均有差异。T2DM患者中AMPK-rs2796516G/A基因多态性与胰岛素抵抗、血脂代谢、血清脂联素及抵抗素水平等明显相关,与G/G、G/A组相比,A/A组HOMA-IR明显增高,TG、TC及LDL增高,血清脂联素水平减低,抵抗素水平增高。结论甘肃地区汉族人群AMPK-rs2796516基因多态性与T2DM及血清脂联素、抵抗素水平有明显相关性。 Objective To investigate the association among AMPKα2 subunit gene polymorphism in Han People from Gansu and type 2 diabetes mellitus (rlEDM), insulin resistance (IR) and plasma resistin and adiponectin. Methods 188 patientes with T2DM(case group) and 179 healthy persons(control group) were enrolled. A SNP rs-2796516( G 〉 A) in AMPKα2 subunit gene was genotyped in all the subjects enrolled using PCR and restriction fragment length polymorphism (RFLP). ELISA technique was employed to determine the plasma adiponectin and resistin concentration. At the same time the fasting blood glucose, insulin and blood lipid were detected. Results There was polymorphism of AMPK-rs2796516 in both T2DM and normal subjects. There was significant difference of genotype constituent ratio and A al.lele frequency in two groups. In case group, AMPKα2 subunit gene polymor- phism was significantly related to insulin resistance, blood lipid and the concentration of plasma adiponectin and re- sistin. Compared with GG and C,.A group, the concentration of serum TG, TC, LDL, resistin were higher in AA group, the concentration of serum adiponectin was lower in AA group. Conclusions The polymorphism of AMPK- rs2796516G/A was revelant with T2DM, the concentration of plasma adiponectin and resistin.
