摘要
为了研究体重和激素重复处理对羔羊卵泡及卵母细胞发育能力的影响,试验采用注射外源FSH+PMSG的方法,对5~15周龄的羔羊进行激素处理,诱导其卵泡发育并活体采卵,卵母细胞体外培养。结果显示:(1)体重居中(M组)的羔羊获得的卵泡数目多于体重较轻(L组)和较重(W组)的羔羊组,但各组之间差异不显著;M组卵母细胞数目多于其他两组,且差异显著(P<0.05)。(2)M组羔羊获得的卵母细胞体外发育卵裂率高于L组和W组,但各组间差异不显著;W组桑囊率高于其他两组,且与L组之间差异显著(P<0.05)。(3)重复处理组与初次处理组相比较,卵巢直径差异不显著,而重复处理组羔羊卵泡数目和获得的卵母细胞数目明显少于初次处理组,差异极显著(P<0.01),同时,重复组获得的卵母细胞卵裂率和桑囊率均低于初次组。结论:羔羊体重居中,其卵母细胞数目和细胞的发育率均好于体重过轻过重的羔羊,重复处理后羔羊所获卵母细胞数目少于初次处理。
The effects of body weight and hormone repeated on lamb oocytes development were studied. The experiments included that : ( 1 ) Three group lambs of different body weight and aged from 5 to 15 weeks were stimulated by multiple dose of FSH and a single injection of PMSG. The oocytes were cultured for observing rate cladvage and blastocyst obtained by laparotomy oocytes recovery from live donors. (2) Lambs of similar body condition were stimulated repeatedly with same hormone, and then the oocytes were cultured. The results were as follows: (1) There was no significant difference among three groups in number of follicles ; but the number of oocytes of group M ( middle weight) were signif- icantly higher than the other two groups (P 〈 0.05 ). (2) The cleavage and mulberry/blastocyst rates of oocytes from Group H (heavy) were higher than those from group M (middle) and group L ( light), and the mulberry/blastocyst rates of oocytes from group H (heavy) was significantly higher than those from group L ( P 〈 0. 05 ). (3) The number of follicles and oocytes from repeat treatment were significantly lower than the initial treatment (P 〈0. O1 ). In conclusion, the number of oocytes and the development capability of oocytes from the middle weight lambs were better than the other two groups. The number of oocytes of the group treated repeatedly was less than initially.
