摘要
研究了林可霉素生物合成基因簇中调控基因lmbU的功能。通过构建重组质粒pLIN-DU和pLIN-U分别对林可霉素工业生产菌株Streptomyces lincolnensis的lmbU基因进行同框敲除和基因倍增,获得相应突变菌LIN-DU和LIN-U后,对突变菌发酵液进行生物效价测定和LC-MS分析,探讨lmbU基因对林可霉素产量和组分的影响。结果发现,基因敲除突变菌LIN-DU的林可霉素生物合成能力大幅削弱;基因倍增突变菌LIN-U的林可霉素生物合成相对出发菌株没有明显变化。表明lmbU基因对林可霉素生物合成具有正向调控作用。
The role of lmbU from the biosynthetic gene cluster of lincomycin in its production was studied.The recombinant plasmid pLIN-DU was constructed by in-frame deletion of lmbU in an industrial lincomycin-producing strain Streptomyces lincolnensis,whereas the plasmid pLIN-U was made to duplicate lmbU there.Lincomycin production had been examined upon bio-assay and LC-MS analysis in the resulting mutants LIN-DU(for lmbU in-frame deletion) and LIN-U(for lmbU duplication) by using that of the original strain as the control.It was found that lincomycin production dramatically decreased in the lmbU-deleted strain LIN-DU but had no significant change in the lmbU-duplicated strain LIN-U,which showed lmbU regulatory gene had a positive effect on lincomycin biosynthesis.
