糖尿病大鼠的肾脏肥大和尿EGF变化 被引量:5

Urinary epidermal growth factor in renal hypertrophy in streptozotocin-diabetic rats

摘要 本文观察了大黄对链脲菌素所致糖尿病大鼠肾脏肥大、尿表皮生长因子(EGF),以及肾功能的影响.结果表明,糖尿病大鼠肾脏肥大和尿EGF呈明显相关性,大黄抑制肾脏肥大,其尿EGF排泄量也明显降低;同时肾小球滤过率降低,大黄并能部分地减少近曲小管Na^+的再吸收.说明大黄抑制肾脏肥大的作用与大黄减少肾EGF生成和近曲小管增殖和肥大有关. Epidermal growth factor(EGF) in urine of 10 streptozotocin(STZ)-diabetic rats and 10 Rheum Officinale(RO), a Chinese herb drug, treated diabetic rats were measured to study the role of EGF in initiating renal hypertrophy in the diabetic rats. Renal hypertrophy occured in the diabetic rats but not in the RO-treated rats. Urine EGF was 38. 2± 16. 6ng/ 24h at day 28 in control, and it was significantly increased from day 7 in the diabetic rats (477. 8± 187. 2ng/24h at day 7, 328. 2± 153. 9ng/24h at day 28), while in the RO-treated rats it was significantly less than that in the diabetic rats(282. 2± 112. 5ng/24 h at day 7,258. 3 ±73. 3ng/24 h at day 28). Moreover, GFR and sodium reabsorption in proximal tubules were reduced in RO-treated diabetic rats as compared with STZ-diabetic rats. Because the kidney is the main source of urine EGF, and EGF has been shown to induce renal growth both in vitro and in vivo,we conclude that EGF may be able to initiate renal hypertrophy in diabetic rats, and effects of RO on the renal hypertrophy is probably relative to the inhibition of renal EGF production resulted by RO treatment.
