摘要
应用放射受体分析法对肝性脑病(HE)及对照大白鼠脑内生长抑素(SS)受体的结合特性进行测定,以探讨SS受体在HE发生中的作用。放射配基用(125)~I—SS,实验数据用Scatchard作图法分析。结果:HE鼠平衡解离常数(Kd)为3.99±0.22nmol/L,最大结合容量(B_(max))为238.0±14.2fmol/mg蛋白质;较对照鼠B_(max)值明显增加(P<0.05)而Kd值无显著差异(P>0.05)。本研究结果表明HE较对照鼠脑内SS受体结合位点数量增加,亲和力不变。提示其在HE发生中具有一定作用。
The present study was undertaken to evaluate the effect of somatostain receptor,a brain-gut peptide receptor which is capable of inhibiting central neurons,in the patho genesis of hepatic encephalopathy(HE).By means of radioligand binding assay,somato statin(SS)receptors in crude synaptosomal membrane of rat brains were investigated in a rat model of HE induced by partial hepatectomy following carbon tetrachloride intoxication and in controls.Binding to SS receptor was studied using^(125)I-SS as radioloigand.Scatchard analysis of binding data was linear,yielding a dissociation constant(Kd) of 3.99±0.22 nmol/L and a maximal binding capacity(B_(max))of 238±14.2 fmol/mg of protein in HE rats.Increased Bm_(max) values were observed(P<0.05)but the Kd values were statistically unchanged(P>0.05)in HE rats as compared with those in controls.The results suggest that the changes of SS receptors in brains play a significant role in the pathogenesis of HE.
