摘要
应用反转录-聚合酶链式反应(ReverseTranscription一PlymeraseChainReaction,RT一PCR)方法,亚克隆出大鼠小脑和肾脏NOScDNA,并以此为探针,通过Northernblotting分析和定量PCR方法,测定大鼠中枢和外周一些组织中NOSmRNA的含量,证明NOSmRNA广泛存在于中枢和外周组织中,在中枢以小脑含量为最高,在外周组织中以肾脏含量为最高;定量PCR方法测定NOSmRNA较Northernblotting分析更为灵敏。
With Reverse
Transcription- Polymerase Chain Reaction(RT- PCR),nitric oxide
synthase(NOS) com-plementary DNA has been subcloned from rat
cerebellum and kidney.The levels of NOSmRNA in some cen-trical and
peripheral tissues in rat have been detectd by Northern blotting and
quantitative PCR.It has been proved that NOS mRNA is existed
extensively in centrical and peripheral tissues.Among of them,NOS mR-
NA Ievels are highest in cerebellum in centre and in kidney in
peripheral tissues.Moreover, quantitative PCR is more sensitive than
Northern blotting in detecting NOS gene expression.
