丹参对体外培养鸡胚胎股骨生长的影响 被引量:2

摘要 实验用孵育9天的鸡胚胎,在无菌条件下取双侧股骨。右侧股骨用1640、Eagle MEM、199和 BGJb 等4种培养基分别进行培养(对照组);左侧股骨用含0.8%丹参注射液的上述4种培养基分别培养(丹参组)。在38℃、CO_2培养箱,用旋转管培养技术分别培养1周、2周和3周。结果发现:(1)体外培养的骨组织由于没有血管的长入,两端没有骨化中心形成。股骨长度的增大只能靠软骨组织的发育和成熟。(2)骨领长度的增加可以通过骨外膜的成骨作用而有所延伸。但这不会导致股骨长度的增加。骨干横径和骨量的增加则有赖于膜内成骨过程。(3)丹参能促进胚胎骨两端的原软骨组织发育和成熟为肥大软骨细胞,并发生钙化;又能促进骨外膜细胞分裂、增殖并逐步分化成为骨细胞,形成骨组织。 The right femora taken from embryonic chickens,which had been hatched for 9 days were put into test tubes con- taining the RPMI 1640,Eagle MEM,199 or BGJb media and cultured in vitro separately as the control groups;while the left femora were cultured in the above- mentioned 4 media with Radix Salviae miltiorrhizae (RSM) added to each of them as the experimental groups.All the test tubes were mounted on a roller drum and cultured in CO_2 incubator for 1,2 or 3 weeks.It was found that:(1) Owing to the fact that no blood vessels grew into the bone tissues cultured on vitro,there was no formation of ossification center at the two ends of the embryonic femora.The increase of femoral length could only depend on the development and maturation of the cartilage tissue;(2) Increase in the length of the embryonic bone collar might be due to intramembranous ossification leading to its extension.But this did not lead to increase in the length of the embryonic femora.The increase in transversal diameter and bone volume of the diaphysis depended on the intramembra- nous ossification;(3) Different types of media had different influences on the embryonic bone tissue formation.If the cul turing period lasted over a week,1640 medium would lead to necrosis and degradation of bone tissue.The growth rate of the cartilage and bone tissue was slow in the Eagle MEM medium.But the rate was rapid in the 199 medium,owing to its full nutrition.BGJb medium was specially designed to maintain the embryonic cartilaginous bone.The types,contents and ratios of the amino acids in BGJb medium met the requirement of bone tissue growth well and the content of calciun and phosphorus in the medium was high.The product of calcium and phosphorus was three times more than that of the above-mentioned three types of media.Consequently the embryonic bone tissue grew very fast in BGJb medium,and calci- fication of bones in this medium occurred the earliest and most extensive,which was manifested not only by an increase in the transversal diameter and volume o
