摘要
目的:研究"双固一通"温针灸在预防兔膝关节骨性关节炎过程中对兔膝关节软骨细胞凋亡的影响。方法:24只日本大耳白兔随机分为"双固一通"温针灸组、模型组和正常组,每组8只。采用结扎股静脉法建立兔膝骨性关节炎模型,"双固一通"温针灸组在造模开始时采用"双固一通"温针灸法,对关元(CV 4)、足三里(ST 36)、内膝眼(EX-LE4)、犊鼻(ST 35)和阳陵泉(GB 34)等穴进行预防性治疗;正常组不做任何处理,正常喂养。在造模8周后,各组分别通过透射电镜观察兔膝关节软骨细胞的超微结构,TUNEL染色光镜下观察软骨细胞凋亡情况。结果:"双固一通"温针灸组兔膝关节软骨细胞超微结构接近正常组,模型组软骨细胞轮廓不清楚,胞膜坏死崩解,胞浆内细胞器消失,细胞裂解为致密颗粒样物质,细胞固缩、变形,甚至出现核分裂,染色质浓聚;"双固一通"温针灸组和模型组软骨细胞凋亡指数高于正常组(P<0.01),"双固一通"温针灸组软骨细胞凋亡指数低于模型组(P<0.01)。结论:"双固一通"温针灸可抑制实验性兔膝骨性关节炎软骨细胞过度凋亡,从而对兔膝骨性关节炎起防治作用。
Objective：To observe the effect of ＂Shuanggu Yitong＂ warming acupuncture on the apoptosis of articular cartilage in rabbits with knee osteoarthritis.Methods： Twenty four healthy Japanese White Rabbits were randmonly divided into three groups：＂Shuanggu Yitong＂ warming acupuncture group,model group and normal group.Model of knee osteoarthritis was established with ligation of femoral vein,and intervention was used at the beginning of model preparation.Animals in the ＂Shuanggu Yitong＂ warming acupuncture group were treated with ＂Shuanggu Yitong＂ warming acupuncture applied on Guanyuan（CV 4）,Zusanli（ST 36）,Neixiyan（EX-LE4）,Dubi（ST 35） and Yanglingquan（GB 34）（each acupoint for 20 minutes,once a day）;animals in the model group with nothing.At eight weeks after model preparation,Ultrastructure of chondrocyte in each group were evaluated through transmission electron microscopic（TEM）,and the apoptosis of articular cartilage was detected by the terminal deoxnucleotidyl transferase-mediated dUTP nick-end labeling（TUNEL） assay.Results： The ultrastructure of chondrocytes in ＂Shuanggu Yitong＂ warming acupuncture group was similar to in normal group.Membranes of chondrocytes in model group were incomplete with several fractures and the cells appeared to leak contents,vacuoles were also seen under the nuclear membrane with bare nuclei,changes were observed with irregular nuclei,prominent vacuoles were observed in early apoptotic cells,obliterated the cytoplasm and its organelle.Apoptosis index（AI） for chondrocytes in ＂Shuanggu Yitong＂ warming acupuncture group and model group was higher than in normal group respectively（P〈0.01）,AI for chondrocytes in ＂Shuanggu Yitong＂ warming acupuncture group was lower than in model group（P〈0.01）.Conclusion： ＂Shuanggu Yitong＂ warming acupuncture can inhibit the apoptosis of cartilage and delay the degeneration of articular cartilage changes,which may contribute the therapeutic effect of warming acupunctur
