佛波醇酯对MES 23.5细胞铁调节蛋白1表达的影响

THE EFFECT OF PMA ON THE ACTIVATION OF IRON REGULATORY PROTEIN-1 IN MES 23.5 CELLS

摘要 目的探讨蛋白激酶C(PKC)激动剂佛波醇酯(PMA)对MES 23.5细胞中铁调节蛋白1(IRP1)的激活作用。方法采用MTT法观察细胞的存活率,Western Blot法检测IRP1和铁转出蛋白(Fpn1)的表达变化。结果 0.01~2.00μmol/L的PMA作用MES 23.5细胞24 h后,细胞存活率无明显变化(F=3.050,P〉0.05)。0.20μmol/L的PMA作用MES 23.5细胞24 h后,IRP1表达明显上调(t=4.501,P〈0.01),Fpn1表达明显下调(t=5.174,P〈0.01)。结论 PMA在多巴胺能神经细胞系MES 23.5细胞中能明显激活IRP1。 Objective To study the activation of phorbol ester,agonist of protein kinase C(PKC) on iron regulatory protein-1(IRP1) in dopaminergic cell line MES 23.5 cells. Methods Cell viability was assessed by MTT.IRP1 and Fpn1 levels were investigated by Western Blot. Results After treatment with 0.01-2.00 μmol/L PMA for 24 hours,the survival of MES23.5 cells was not changed(F=3.050,P0.05).After treatment with 0.20 μmol/L PMA for 24 hours,the expression of IRP1 upregulated(t=4.501,P0.01),and Fpn1 expression down-regulated(t=5.174,P0.01). Conclusion IRP1 can be activated by PMA in dopaminergic MES 23.5 cells.
