摘要
目的对灭菌物品进行分枝杆菌菌种鉴定,确定灭菌物品是否有分枝杆菌,以达到分枝杆菌卫生监测。方法无菌生理盐水拭子法擦洗灭菌物品,用TritonX-100裂解液提取DNA,采用16s-23s rDNA PCR法,扩增特定间区片段,琼脂凝胶电泳进行分型鉴定。结果 200份有效期内的灭菌物品扩增,没有分枝杆菌特异带;200份过期的灭菌物品扩增,有1份物品扩增出草分枝杆菌异常带;阳性对照的标准菌株出现特定扩增,阴性对照没有扩增带。结论通过16s-23s rDNA PCR法可以对灭菌物品进行分枝杆菌监测。
Objective To identify mycobacteria species on sterilized items.Methods The steilzied items were cleaned with saline and the TritonX-100 lysis buffer was used to extract bacterium DNA by using 16s-23s r DNA PCR method.The specific fragment was amplified and determined with agar gel electrophoresis for species typing and identification.Results There no mycobacterium were idendified from 200 sterilized items within the validity period,while a specific band was observed from another 200 sterilized out-of-date items.Conclusions 16s-23s r DNA PCR method can be used for monitrong contamination of sterilized items with mycobacterium.
