摘要
目的观察间充质干细胞(mesenchymal stem cells,MSC)在大鼠异种肝移植肝组织中的迁徙、定居及组织修复作用。方法分离培养、体外扩增豚鼠骨髓MSC,经4,6-联脒-2-苯基吲哚(4,6-diamidino-2-phenylindole,DAPI)标记后用荧光显微镜进行体外示踪观察。制作豚鼠对Wistar大鼠的肝移植模型,DAPI标记的MSC经股静脉植入Wistar大鼠,用荧光显微镜观察术后12h、24hMSC在移植肝的迁徙及定居情况;采用苏木素-伊红(HE)染色观察移植肝组织的损伤情况。结果 DAPI标记MSC的标记率达100%,标记后MSC生长和增殖特性未受影响。MSC经股静脉注入后,12h即出现在移植肝,24h分布于移植肝血管及周围组织。显微镜下观察,空白对照组术后24h的肝小叶中央静脉和肝血窦淤血明显,肝细胞可见严重空泡状变性,坏死灶较多,肝组织中有较多中性粒细胞的聚集、浸润;MSC组肝小叶、中央静脉、汇管区结构清晰,炎症细胞浸润较轻。结论 MSC可以迁徙并定居于大鼠异种移植肝中,对移植肝组织损伤有修复作用。
Objective To observe the effect of mesenchymal stem cells (MSC)on metaptosis, settlement and tissue repair of hepatic tissue in rat heterogenous liver transplantation. Methods Bone marrow MSC of guinea pig were separated, cultured and expanded in vitro, and were labeled with 4, 6-diamidino-2-phenylindole (DAPI) and traced by fluorescence microscope in vitro. The model of liver transplantation from guinea pig to Wistar rat was established. The DAPI labeling MSC were transplanted into Wistar rat through femoral vein. The metaptosis and settlement of MSC in transplant liver were traced at 12 h, 24 h after operation by fluorescence microscope. The tissue injury of transplant liver was observed by hematoxylin-eosin (HE) staining. Results The rate of DAPI labeling MSC was 100 %. The growth and proliferation of MSC did not change after DAPI labeling. The DAPI labeling MSC were found around the transplant liver at 12 h after injected through femoral vein and were found around the vessel and tissue of transplant liver at 24 h after injection. Under the microscope, passive congestion was found in acini hepatis central vein and hepatic sinusoid in control group at 24 h after operation and serious cavity denaturation and necrosis focus were found in hepatic cells, and neutrophile granulocyte aggregation and infiltration were found in hepatic tissue. In MSC group, the clear structure was seen in acini hepatis, central vein and portal area with mild inflammatory cell infiltration. Conclusion MSC can migrate and settle in the transplant liver of rat and is helpful to repair the injured tissue of transplant liver.
