摘要
目的:探讨在反式-7,8-二羟-9,10-环氧苯并芘(anti-BPDE)诱导转化的人支气管上皮细胞(16HBE-T)中低表达的mir-493-5p在化学致癌物致癌过程中的作用。方法:运用实时荧光定量PCR(qRT-PCR)验证miR-493-5p成熟体在16HBE-T和非转化对照细胞(16HBE-N)的表达水平。瞬时转染miRNA的模拟物,改变16HBE-T中miR-493-5p的表达,检测转染后16HBE-T细胞的增殖能力。结果:16HBE-N细胞中mir-493-5p的表达水平是16HBE-T的0.28倍,在16HBE-T组过表达mir-493-5p后实验组细胞增殖能力下降。结论:mir-493-5p表达水平的改变对细胞的增殖能力有影响,在anti-BPDE恶性转化细胞中低表达的mir-493-5p可能发挥着类抑癌基因的作用。
Object:To explore the function of mir-493-5p which is low expressed in the anti-benzo[a]pyrene-7,8-diol-9,10-epoxide(anti-BPDE) induced transformation of human bronchial epithelial cells (16HBE-T) and the role of mir-493-5p in the chemical induced Cancers' process. Methods:The relative expression level of mature mir-493-5p in 16HBE-T and non-transformed control cells (16HBE-N) was measured by real-time quantitative reverse transcription polymerase chain reaction (qRT-PCR) .We transient transfect miR-493-5p' mimics to upregulate the expression in 16HBE-T. Detected the cell proliferation after transfecting 16HBE-T. Results:0.28 - fold expression level change of mir-493-5p was observed in the 16HBE-N cells compaired with 16HBE-T cells before transfection. Over-express mir-493-5p induced the experimental group decreased cell proliferation. Conclusion:The change of expression of mature mir-493-5p in 16HBE-T cells can affect the cell proliferation.Mir-493-5p with a lower expression in 16HBE-T may be a likeness anti-oncogene.
