摘要
目的:研究间歇性应用重组人甲状旁腺素[rhPTH(1-34)]对兔下颌骨骨折的治疗作用。方法:健康新西兰大白兔24只,体重1.9~2.1kg。在下颌骨体部造成1.5mm×10mm的部分骨折模型,随机分成A、B 2组,每组12只(雌雄不限)。A组分别于术后第2天间隔天数行术区局部rhPTH(1-34)30μg/(kg.d)注射,B组不做处理。于术后不同时间点行X线片、骨密度、HE染色和免疫组织化学染色观察,以及测定血清钙素水平。结果:术后20d各组骨密度达峰值,A组骨密度高于B组(P<0.05)。术后A组骨折愈合处colⅡ蛋白分泌较B组增多(P<0.05)。术后7、20及30d各组血清骨钙素水平均较对照组明显增高(P<0.05)。结论:以30μg/(kg.d)剂量注射rhPTH(1-34)可安全有效促进兔下颌骨骨折模型骨折愈合。
Objective: To detect the influence of rhPTH(1-34) on mandibular fracture healing in rabbit.Methods: Twenty-four healthy New Zealand white rabbits weighing 1.9-2.1 kg were used.Which were caused the mandibular body 1.5x10mm part fracture,and randomly divided into two groups with 12 in each group(male or female).Group A from the second postoperative day were injected the local rhPTH(1-34) 30μg /(kg·d).Group B did not deal with.Lines at different time points were measured the bone mineral density,observed X-ray film,immuneohistochemical staining,and serum calcium levels.Results: Bone mineral density after 20 d in each group reached the peak.Group A bone mineral density was higher than that of group B(P0.05).The colⅡ protein secretion at fracture healing after surgery of group A was higher than group B(P0.05).After 7,20 and 30 days the serum osteocalcin levels were significantly higher than those in the control group(P0.05).Conclusion: 30μg /(kg·d) dose injection of rhPTH(1-34) can effectively promote the safety mandibular fracture healing of fracture rabbit model.
