狼尾蕨叶片诱导不定芽植株再生技术 被引量:4

摘要 以狼尾蕨无菌苗的叶片为外植体,探讨不同类型的细胞分裂素对不定芽诱导、增殖和成苗的影响。结果显示:在附加1.0 mg/L和2.0 mg/L KT或1.0 mg/L和2.0 mg/L CPPU的诱导培养基上叶片诱导产生叶状体状不定芽,转到添加0.5 mg/L KT或0.5 mg/L CPPU的增殖培养基上可形成叶芽状不定芽;在附加1.0 mg/L和2.0mg/L BAP的培养基上叶片诱导可产生叶状体状不定芽,转到添加0.5 mg/L BAP的增殖培养基上不定芽仍保持叶状体状。叶芽状不定芽在附加0.5 mg/L CPPU的增殖培养基上连续继代培养,每代芽增殖系数达5以上。叶芽状不定芽转到附加0.2 mg/L IBA的成苗和生根培养基上,能发育成具有根、茎、叶的完整小植株,成苗率达100%。 The leaves of sterile plantlet of Davallia bullata was used as explant to study the effect of different types of cytokinin on adventitious bud induction,proliferation and shooting.The thallus-like adventitious bud was induced from the leaf in the modified MS medium with 1.0mg/L KT,or 2.0 mg/L KT,or 1.0 mg/L CPPU,or 2.0mg/L CPPU,and formed leafbud-like adventitious bud in the bud proliferative medium with 0.5 mg/L KT or 0.5 mg/L CPPU.The thallus-like adventitious bud was induced as well from the leaf in the induction medium with 1.0 mg/L or 2.0 mg/L BAP and stayed the same shape in proliferation medium with 0.5 mg/L BAP.In the bud proliferative medium with 0.5 mg/L CPPU,the coefficient of bud propagation was above 5.The leafbud was 100%,and developed into intact regenerated plantlet with root,stem and leaf in the MS medium with 0.2 mg/L IBA.
