基于线粒体COI基因部分序列分析4种松毛虫的亲缘关系 被引量:4

摘要 为了探讨4种松毛虫的亲缘关系,对马尾松毛虫、油松毛虫、赤松毛虫和落叶松毛虫的线粒体COI基因序列进行了扩增和测序。在获得的636 bp的COI序列中碱基的平均频率为:A=34.75%,T/U=34.75%,C=15.25%,G=15.25%。马尾松毛虫遗传多样性最高(核酸多样度为3.93%),落叶松毛虫最低(0.629%)。4种松毛虫间的亲缘关系由远到近依次为:落叶松毛虫×赤松毛虫(0.083)、落叶松毛虫×马尾松毛虫(0.068)、落叶松毛虫×油松毛虫(0.066)、赤松毛虫×油松毛虫(0.057)、马尾松毛虫×赤松毛虫(0.054)、马尾松毛虫×油松毛虫(0.012)。基于线粒体COI基因序列能够鉴别4种松毛虫之间的亲缘关系,可作为种上的分子分类标记应用于松毛虫属昆虫系统发育的研究。 Genetic differentiation among four species of Dendrolimus punctatus , D. tabulaeformis , D. spectabilis , and D. superans was analyzed based on COI gene sequences in order to clarify the taxonomic status of the four pine moths. The average base frequencies of636bp COI gene sequences were obtained as A 34.75%, T/U 34.75%, C 15.25%, and G 15. 25%. D. punctatus has the lfighest genetic diversity (3.93%), while file genetic diversity of D. superans is the lowest (0. 629% ). The genetic distance among the four spices is in the order of D. superans × D. spectabilis (0. 083 ) , D. superans × D. punctatus (0.068), D. superarts x D. tabulaeformis (0.066), D. speetabilis × D. tabulaeformis (0.057), D. punctatus × D. spectabilis (0.054), and D. punctatus x D. tabulaeformis (0.012). The COI sequence can be used to identify the genetic relationship between the four species of pine months and is a suitable marker in Dendrolimus molecular taxonomy.
